Nuclear Tracks in Solids (Principles and Applications)

Author(s): Chen, Fanqing; Gerion, Daniele | Abstract: One of the biggest challenges in cell biology is the imaging of living cells. For this purpose, the most commonly used visualization tool is fluorescent markers. However, conventional labels, such as organic fluorescent dyes or green fluorescent proteins (GFP), lack the photostability to allow the tracking of cellular events that happen over minutes to days. In addition, they are either toxic to cells (dyes), or difficult to construct and manipulate (GFP). We report here the use of a new class of fluorescent labels, silanized CdSe/ZnS nanocrystal-peptide conjugates, for imaging the nuclei of living cells. CdSe/ZnS nanocrystals, or so called quantum dots (qdots), are extremely photostable, and have been used extensively in cellular imaging of fixed cells. However, most of the studies about living cells so far have been concerned only with particle entry into the cytoplasm or the localization of receptors on the cell membrane. Specific targeting of qdots to the nucleus of living cells has not been reported in previous studies, due to the lack of a targeting mechanism and proper particle size. Here we demonstrate for the first time the construction of a CdSe/ZnS nanocrystal-peptide conjugate that carries the SV40 large T antigen nuclear localization signal (NLS), and the transfection of the complex into living cells. By a novel adaptation of commonly used cell transfection techniques for qdots, we were able to introduce and retain the NLS-qdots conjugate in living cells for up to a week without detectable negative cellular effects. Moreover, we can visualize the movement of the CdSe/ZnS nanocrystal-peptide conjugates from cytoplasm to the nucleus, and the accumulation of the complex in the cell nucleus, over a long observation time period. This report opens the door for using qdots to visualize long-term biological events that happen in the cell nucleus, and provides a new nontoxic, long-term imaging platform for cell nuclear processes.

Ammonia gas sensors: A comprehensive review.

We report the use of laser-induced breakdown spectroscopy (LIBS) for determination of soil contamination due to effluents from the leather tanning industry in Kasur district of Punjab in Pakistan. The quantitative analysis was performed by making a calibration curve for Cr using LIBS spectral line at 427.48 nm. Chromium concentration of 839 mg·kg -1 in the vicinity of an effluent drain and 1,829 mg·kg -1 in the area of an old stagnant pool was found. The leaching of Cr due to seepage of industrial effluents from deteriorating bricklined drains in horizontal direction also was observed.

Chromium Quantification in a Leather Tanning Industrial Area Using Laser Induced Breakdown Spectroscopy

Magnetic nanoparticles have been extensively in the biomedical field as drug delivery agent, diagnosis of different diseases and more recently in the treatment of different types of cancer. Majority of these studies reported the use of iron oxide nanoparticles or formulation contains at least iron in most of magnetic-based nanosystems. Focus on iron oxide nanoparticles is mainly due to their superparamagnetic nature at nanoscale and other features such as higher surface to volume ratio, biocompatibility and low toxicity. For further improvement in their properties, doping with different transition metal elements is also under investigations. This chapter covers most commonly used types of iron oxide NPs, behavior with few doping metals in iron oxide nanoparticles and their synthesis protocols through different physical and chemical methods. Finally, heat generation mechanisms responsible for localized heat in tissues have been discussed.

Thermal Response of Iron Oxide and Metal-Based Iron Oxide Nanoparticles for Magnetic Hyperthermia

The present study was conducted to assess the effects of different fluences of Nd:YAG pulsed laser irradiation on the kernel thermodynamic attributes and biophoton emission in relation with the changes in kernel metabolites during kernel germination. Significant changes in kernel internal energy, enthalpy change, entropy generation, entropy generation ratio, entropy flux, entropy flux ratio, and biophoton emission were observed. Lower laser fluences (200, 400 and 600 J/cm2) positively influenced these attributes in association with a rapid increase in kernel chlorophyll (Chl.), carotenoids (Car.), total soluble sugar (TSS), total reducing sugar (TRS), total free amino acids (TFAA) and free fatty acids (FFA) contents as compared to non-treated ones but the opposite was true at higher fluences (800 J/cm2 and above). Correlations and PCA analysis were performed for the studied attributes that showed strong negative correlations of kernel Chl., Car., TSS, TRS, TFAA and FFA with kernel TSP and oil contents that confer the breakdown of kernel reserves and this conversion was faster at low laser fluences. In conclusion, 200 and 400 J/cm2 fluences have been found the most beneficial ones as kernel treatment in maize for better metabolic activities to improve the germination.

A Comprehensive Study on Pulsed Nd:YAG Laser Irradiation of Maize Kernels: Measurement of Biophoton Emissions, Metabolite Levels and Thermodynamical Attributes During Germination

Developing contamination-free techniques for synthesizing nanostructures, especially bimetallic nanoparticles for hyperthermia applications has gained more attention in recent years. Au/Co nanoparticles (NPs) were synthesized using pulsed Nd:YAG laser ablation in distilled water due to its simple, rapid, environmentally friendly, and contamination-free nature. Transmission Electron Microscopy (TEM) analysis showed that the synthesized nanoparticles were spherical in shape and 15 nm in size. The phase structure of the synthesized NPs was examined using X-ray diffraction (XRD). UV–visible spectrometer and vibrating sample magnetometer (VSM) techniques were used to study the optical and magnetic properties of the synthesized materials. Au/Co nanoparticles showed non-mutagenic potential against S. Typhimurium TA-98 and S. Typhimurium TA-100 strains. Furthermore, a lab-made apparatus was used to study the magnetic hyperthermia of Au/Co nanofluid. The specific absorption rate (SAR) of 4.4 W/g was estimated for magnetic field strength of 177 μT. The study shows that the synthesized nanoparticles have magnetic hyperthermia potential.
