Graphene-based materials for tissue engineering.

Functional graphene nanomaterials (FGNs) are fast emerging materials with extremely unique physical and chemical properties and physiological ability to interfere and/or interact with bioorganisms; as a result, FGNs present manifold possibilities for diverse biological applications. Beyond their use in drug/gene delivery, phototherapy, and bioimaging, recent studies have revealed that FGNs can significantly promote interfacial biointeractions, in particular, with proteins, mammalian cells/stem cells, and microbials. FGNs can adsorb and concentrate nutrition factors including proteins from physiological media. This accelerates the formation of extracellular matrix, which eventually promotes cell colonization by providing a more beneficial microenvironment for cell adhesion and growth. Functional Graphene Nanomaterials Based Architectures: Biointeractions, Fabrications, and Emerging Biological Applications

Graphene and its derivatives are heralded as ‘miracle’ materials with manifold applications in different sectors of society from electronics to energy storage to medicine. The increasing exploitation of graphene-based materials (GBMs) necessitates a comprehensive evaluation of the potential impact of these materials on human health and the environment. Here we discuss synthesis and characterization of GBMs as well as human and environmental hazard assessment of GBMs using in vitro and in vivo model systems with the aim to understand the properties that underlie the biological effects of these materials; not all GBMs are alike, and it is essential that we disentangle the structure-activity relationships for this class of materials.

Safety assessment of graphene-based materials: focus on human health and the environment

Since the isolation of graphene in 2004, there has been an exponentially growing number of reports on layered two-dimensional (2D) materials for applications ranging from protective coatings to biochemical sensing. Due to the exceptional, and often tunable, electrical, optical, electrochemical, and physical properties of these materials, they can serve as the active sensing element or a supporting substrate for diverse healthcare applications. In this review, we provide a survey of the recent reports on the applications of 2D materials in biosensing and other emerging healthcare areas, ranging from wearable technologies to optogenetics to neural interfacing. Specifically, this review provides (i) a holistic evaluation of relevant material properties across a wide range of 2D systems, (ii) a comparison of 2D material-based biosensors to the state-of-the-art, (iii) relevant material synthesis approaches specifically reported for healthcare applications, and (iv) the technological considerations to facilitate mass production and commercialization.

Two-Dimensional Materials in Biosensing and Healthcare: From In Vitro Diagnostics to Optogenetics and Beyond

Graphene oxides with different surface charges were fabricated from carboxylated graphene oxide by chemical modification with amino- (-NH2), poly-m-aminobenzene sulfonic acid- (-NH2/-SO3H), or methoxyl- (-OCH3) terminated functional groups. The chemically functionalized graphene oxides and the carboxylated graphene oxide were characterized by infrared spectroscopy, X-ray photoelectron spectroscopy, UV-Vis spectrometry, ζ potential measurements, field emission scanning electron microscopy, and contact angle analyses. Subsequently, the resulting graphene oxides were used as substrates for culturing primary rat hippocampal neurons to investigate neurite outgrowth and branching. The morphological features of neurons that directly reflect their potential capability in synaptic transmission were characterized. The results demonstrate that the chemical properties of graphene oxide can be systematically modified by attaching different functional groups that confer known characteristics to the substrate. By manipulating the charge carried by the functionalized graphene oxides, the outgrowth and branching of neuronal processes can be controlled. Compared with neutral, zwitterionic, or negatively charged graphene oxides, positively charged graphene oxide was found to be more beneficial for neurite outgrowth and branching. The ability to chemically modify graphene oxide to control neurite outgrowth could be implemented clinically, especially in cases wherein long-term presence of outgrowth modulation is necessary.

The supramolecular interaction between the host molecule β-cyclodextrin (β-CD) and the guest molecule adamantane is extensively applied in several disciplines. However, recent studies on the applications of this molecular recognition mainly focused on glass, silicon, and gold substrates. Few studies have explored the field of poly(dimethylsiloxane) (PDMS), an optically transparent elastomer widely used in biological microfluidic devices. In the present study, various functional group-modified adamantyl-terminated dendronized molecules were synthesized via an efficient and facile route. The binding of adamantyl-terminated dendronized molecules onto β-CD-conjugated PDMS surfaces and their reversible dissociation from PDMS surfaces through the competitive mechanism of β-CD-N3 were studied. The results showed that the dissociation is relatively slow compared with the binding, and the former process can be accelerated through exchanging the original β-CD-N3 solution with a fresh one. Afterward, more complex multilayer assemblies were constructed on PDMS surfaces using the host–guest interaction between adamantane and β-CD as well as between biotin and streptavidin. A further study on the association and dissociation of the established multilayer assemblies showed that the assembly constructed with a biotinylated monoadamantyl-terminated molecule has a reversible property, whereas that with a biotinylated triadamantyl-terminated molecule has an irreversible property. These results demonstrated that the reversible properties of the fabricated assembly can be easily modulated on the PDMS surfaces by regulating the valence of the supramolecular interaction between adamantane and β-CD.
