Abstract: Perisperm starch granules of the dicotyledonous plant Amaranthus hypochondriacus L were prepared from two homozygous lines (WxWx and wxwx) and their hybrid (Wxwx) The hybrid line was obtained by natural hybridization By Sephadex G-75 column chromatography of isoamylase-debranched starches, the amylose content of WxWx starch was 169%, that of Wxwx was 107, and wxwx was zero SDS-polyacrylamide gel electrophoresis showed that starch granules from two genotypes (WxWx and Wxwx) contained a Wx protein (MW = 68,000) which was supposed to be a starch granule-bound starch synthase and was associated with amylose synthesis, as observed in nonwaxy maize The intensities of the stained protein bands were apparently correlated with the number of the Wx alleles The Wx protein was not detected in the wxwx starch These findings suggest that the Wx allele produces the Wx protein and amylose in the perisperm of A hypochondriacus, with incomplete dominance over the wx allele The Wx allele did not affect the fine s

Abstract: Seeds of grain amaranths contain a high amount (about 60% of total nitrogen) of albumin and globulin and a trace amount of prolamin. From salt-soluble extracts of A. hypochondriacus seeds, a globulin (440, 000 in apparent molecular weight and s020, w= 12-7) was purified by Sepharose 6B gel and DEAE-cellulose column chromatographies. The protein comprised at least four kinds of subunits whose molecular weights were 36, 000, 32, 000, 20, 000 and 18, 000, respectively. The amino acid composition of the globulin was almost similar to those of soybean and oat globulins.

Abstract: Trypsin inhibitors were isolated from seeds of Amaranthus hypochondriacus by extraction at pH 7.5, heat treatment at 70°C for 15 min and affinity chromatography with trypsin-Sepharose 4B. Inhibitors of trypsin and chymotrypsin were identified in polyacrylamide gels following electrophoresis using a stain procedure that employed the chromogenic substrate acetyl-D, L-phenylalanine-2-napthyl ester. Three of the thirteen trypsin inhibitors identified in the electrophoresis gels, also had antichymotryptic activity. The inhibitor preparation was very thermostable retaining 20% of its original activity after 7 hr at 100°C.
