Abstract: We have studied the mechanism of recombination between human adenovirus DNA and the mammalian genome in considerable detail. Human adenovirus type 2 (Ad2) and type 12 (Ad12) have been investigated. This research was conducted as a model, as it were, to elucidate the mechanism of insertion of foreign DNA into the mammalian genome. There is no doubt that practically any foreign DNA molecule can be taken up by mammalian cells and can be inserted as a whole or in part into the host genome. It is not clear yet how frequent these integration events are. Insertion could be transient in certain cases, and stable integration might be a relatively rare event. As often in molecular biology of eukaryotes, adenoviruses have served a very useful function in the investigation of reaction mechanisms (for recent reviews see 1–3). At least certain aspects of the mechanism of insertion of foreign (viral) DNA into the mammalian genome can be investigated by studying the modes of integration of adenovirus DNA.
