Abstract: The wild herb Swertia mussotii is a source of the anti-hepatitis compounds swertiamarin, mangiferin and gentiopicroside. Its over-exploitation has raised the priority of producing these compounds heterologously. Somatic hybridization represents a novel approach for introgressing Swertia mussotii genes into a less endangered species. Protoplasts derived from calli of Bupleurum scorzonerifolium and S. mussotii were fused to produce 194 putative hybrid cell lines, of which three (all derived from fusions where the S. mussotii protoplasts were pre-treated for 30 s with UV light) later differentiated into green plants. The hybridity of the calli was confirmed by a combination of isozyme, RAPD and chromosomal analysis. The hybrid calli genomes were predominantly B. scorzonerifolium. GISH analysis of mitotic chromosomes confirmed that the irradiation of donor protoplasts increased the frequency of chromosome elimination and fragmentation. RFLP analysis of organellar DNA revealed that mitochondrial and chloroplast DNA of both parents coexisted and recombined in some hybrid cell lines. Some of the hybrid calli contained SmG10H from donor, and produced swertiamarin, mangiferin and certain volatile compounds characteristic of S. mussotii. The expression of SmG10H (geraniol 10-hydroxylase) was associated with the heterologous accumulation of swertiamarin. Somatic hybrids between B. scorzonerifolium and S. mussotii were obtained, hybrids selected all contained introgressed nuclear and cytoplasmic DNA from S. mussotii; and some produced more mangiferin than the donor itself. The introgression of SmG10H was necessary for the accumulation of swertiamarin.

Abstract: Swertia chirayita is a highly traded medicinal plant of Nepal widely used for its anti-diabetic potential. In this study, two herbs (Swertia nervosa and Andrographis paniculata) often used as adulterants and substitutes of Swertia chirayita were analyzed for their antioxidant activity, a-glucosidase inhibitory potential and total phenolic content and compared with that of Swertia chirayita. Aqueous and 12% ethanolic extracts of the three herbs showed moderate to high antioxidant activity and moderate a-glucosidase inhibitory potential. HPLC/ DAD revealed the presence of swertiamarin and mangiferin in all the Swertia species, while Andrographis paniculata contained cinnamates such as cinnamic acid, p-coumaric acid and chlorogenic acid. The antioxidant activity and a-glucosidase inhibitory potential was evident in the two herbs indicating their relevance as substitutes for Swertia chirayita for potential early stage management of type-2 diabetes and related complications.

Abstract: Three new secoiridoids, swertiakoulactone (1) and swertiakosides A and B (2 and 3), were isolated from Swertia kouitchensis. Their structures were elucidated by comprehensive spectroscopic analyses including MS, IR, 1D and 2D NMR data. By the anti-hepatitis B virus (HBV) assay on Hep G 2.2.15 cells line in vitro, compound 1 showed moderate activities inhibiting the HBsAg secretion (IC50 = 1.10 mM, SI = 4.39) and HBV DNA replication (IC50 = 1.16 mM, SI = 4.12). Abstract: As a continuation of our studies on chemical constituents of Swertia plant,from the over-ground parts of Swertia mussotii Franch which is a traditional Tibetan medicine,eight compounds,1,2,6-Trimethoxy-8-hydroxyxanthone(1),1,3-Dimethoxy-8-hydroxyxanthone(2),(z)-5-Ethylidene-3,4,5,6-tetrahydro-cis-6,8-dimethoxy-1H,8H-pyrano-pyran-1-one(3),swertiamarin(4),gentiopicroside(5),Sweroside(6),Stigmasterol(7) and β-sitosterol(8),were isolated.Their structures were determined by using conventional spectroscopic methods.Compound 3 was isolated from the genus for the first time.The effect of compound 1 on aldose reductase was 18.3%(10-5 mol/L).

Abstract: In the given study we had worked on the antidiabetic activity of plant Swerti a chirayita Buch - Ham family Gentinaceae. The ethanolic extra ct was orally administered with the dose of 250mg/kg and 500mg/kg on STZ - NAD induced diabetic albino mice . Findings of this research showed that ethanolic extract possess significant ( P <0.01 ) anti diabetic activity and had beneficial effect on cholesterol and triglyceride level. The results were compared with standard drug Metformin .

Abstract: Swertia species belong to the family Gentianaceae are annual medicinally important herbs, due to presence of xanthone derivatives it has several type of biological activities. Several HPLC methods are used for the quality control of Swertia. The manuscript aims at drawing researchers, scientists' and pharmaceutical companies attention to the HPLC fingerprint of highly valuable medicinal plant Swertia. In most of published papers, xanthones were separated using C18 column as a Stationary phase, methanol and water with or without acid as mobile phase at 1.0 ml/min flow rate. Di fferent detectors viz UV, DAD, PAD, ESI and MS were used for detection.

Abstract: Swertia minor (Griseb.) Knobl. (Gentianaceae) is an endemic, endangered and unexploited medicinal plant found in the regions of the Nilgiris. The present study was designed to evaluate the antidiabetic and hypolipidaemic effect of methanolic extract of Swertia minor in normal and Alloxan induced diabetic rats. After the treatment, a significant reduction was observed in fasting plasma glucose levels, Urea, Creatinine in the diabetic rats. MESM treated rats were showed that considerable depletion of serum total cholesterol, triglycerides, TC/LDL-C and increase in HDL cholesterol. The results were represented as the mean ± SD and the statistical significance was set up at p<0.05 shows that the MESM possess hypoglycemic and hypolipidemic properties.

Abstract: Oleanolic acid (OA) and ursolic acid (UA) are the ubiquitous triterpenic acids in plant kingdom and have multiple biological effects. In the present study, a highly sensitive and selective method using a dual-sensitive probe 2-(5-Benzoacridine)ethyl-p-toluenesulfonate (BAETS) as pre-column labeling reagent has been developed for rapid determination of the triterpenes OA and UA by HPLC with fluorescence detection (FLD) and online mass spectrometry identification. Response surface methodology as an efficient tool was employed to optimize the ultrasonic-assisted extraction of triterpenic acids from Swertia plants and the pre-column derivatization reaction, respectively, which ensured the highest triterpenic acids recoveries within the shortest extraction time and the sufficient labeling of the analyzed components. Fast separation of the isomers OA and UA could be achieved on a Hypersil BDS C8 column within 7 min. Both of OA and UA gave the good correlation coefficients of 0.9999. This developed method offered the satisfactory detection limits of 1.10 and 1.30 ng mL-1 for UA and OA, respectively. When applied to Swertia species, it showed good reproducibility.

Abstract: The genus Swertia Linn. consist of annual and perennial herbs. It comprises 170 species. The medicinal usage of Swertia chirata is reported in treatment of fever, killing worms, regulating blood sugar level, appetizer and in skin diseases. Its medicinal properties are mainly due to the presence of gentian. This article highlighted the ethno botanical, phytochemistry and pharmacological use of the plant Swertia chirata.

Abstract: Antioxidant potential of ethanolic extract of Swertia quartiniana leaves was determined using thiobarbituric acid reactive species (TBARS) and iron chelation assays. The result of the study revealed that incubating rat’s brain and liver homogenates in the presence of 60mMFe2+ caused increased in malondaldehyde (MDA) production in rat’s brain and liver homogenate when compared with normal. However the ethanolic extract of Swertia quartiniana leaves caused decrease in MDA content in both brain and liver homogenates. The extract had higher inhibitory effect at concentration of 29mg/ml (1c50 2.97mg/ml) in the liver and at concentration of 44mg/ml (1c50 0.11mg/ml) in the brain. The extract also showed chelating ability of 76% at concentration of 110mg/ml. The inhibitory effect of the ethanolic extract of Swertia quartiniana using both assays were concentration dependent. It was therefore concluded that ethanolic extract of Swertia quartiniana prevent Fe2+ -induced lipid peroxidation due to higher Fe2+ chelating ability.
