Abstract: Initiation of larval growth, induction of cell necrosis, and gall formation in the host were measured as criteria of resistance or susceptibility of tomato seedlings to the root-knot nematode, Meloidogyne incognita (Kofoid &White) Chitwood. Seedlings grown at 27 C on water agar containing additions were scored 3 or 4 days after infection.In the absence of exogenous plant growth regulatory substances, approximately 73% of larvae that entered roots of susceptible plants showed growth, none induced necrosis and nearly all induced gall formation. In roots of a resistant variety, only 4% of the larvae grew, 88% induced necrosis of host cells, and only 29% induced galls. Exogenously supplied cytokinins shifted the response of the resistant plants toward the susceptible reaction. Exogenous kinetin at 0.4 and 0.8 micromolar allowed 55 and 57% of the nematodes to grow, reduced the incidence of necrosis to 32 and 31%, and increased gall formation to 73 and 65%. Three additional cytokinins, Zeatin, 6-(gamma,gamma-dimethylallylamino)purine, and 6-benzylaminopurine produced effects similar to kinetin. Exogenous indoleacetic acid, gibberellic acid, adenine, guanine, uracil, thymine, cytidine, and 6-methylaminopurine neither increased the percentage of larvae which grew nor decreased the extent of host cell necrosis.

Abstract: The cytokinin activities of extracts of organs developed from the apple fruit bud were compared using the carrot phloem bioassay, and the identity of the cytokinins in the apple fruitlet was investigated. The activity of apple fruitlet extracts was slightly greater than the activity of pedicel extracts, and considerably greater than the activities of extracts of other organs. Extracts of the developing seeds of fruitlets were much more active than extracts of fruitlet flesh. Apple fruitlet extracts contained three principal cytokinins. One was identified as a 6-(substituted amino)purine and was either zeatin or some very closely related compound. The two other cytokinins exhibited the chromatographic behaviour of zeatin riboside and zeatin ribotide. A cytokinin extracted from vegetative apple shoots was chromatographically indistinguishable from zeatin.

Abstract: Compounds related to dihydrozeatin that define the influence of the location of the hydroxyl group along the side chain have been synthesized and tested for cytokinin activity. The compounds compared are in the series: 6-(X-hydroxy-3-methylbutylamino)purines and their ribosides, where X = 2, 3, and 4. Hydroxy substitution on the 4-position of the side chain enhances, but in the 2-, 3-, or 2- and 3- positions, decreases cytokinin activity as compared with the unsubstituted isopentyl (or isopentenyl) chains. This differential influence of the position of the hydroxyl group in the N6-chain holds also for the similarly related 9-β-D-ribofuranosides. The relatively higher activity of 3,4-dihydroxy as compared with 2,3-dihydroxy derivatives is consistent with this position effect. Compounds related to zeatin possessing side-chain ester moieties have also been synthesized and tested comparatively. Among these, 6-(4-acetoxy-3-methyl-trans-2-butenylamino)purine is at least as active as zeatin, the most active presently known cytokinin in the tobacco bioassay, whereas the analog, methyl 2-methyl-4-(purin-6-ylamino)-trans-crotonate, with the ester function effectively reversed, has vastly lower activity, and its riboside is practically inactive.

Abstract: A 6-(gamma,gamma-dimethylallylamino) purine-like compound was found in the culture medium of Rhizopogon roseolus, which had been shown earlier to synthesize zeatin. The role of 6-(gamma,gamma-dimethylallylamino) purine as a precursor of zeatin was studied. Rhizopogon was furnished with 6-(gamma,gamma-dimethylallylamino) purine-8-(14)C. Cochromatography, oxidation studies with potassium permanganate, and bromination indicated that labeled zeatin ribonucleoside was isolated from the medium. The fungus also incorporated labeled adenine, hypoxanthine, and 4-amino-5-imidazole carboxamide into zeatin ribonucleoside.

Abstract: A strain of soybean cells capable of growing on a tissue culture medium lacking a cytokinin produced at least 3 compounds active in the soybean cytokinin assay. The characteristics of these compounds were consistent with their being zeatin in the free form, zeatin ribonucleoside and zeatin ribonucleotide. Abstract: 6-(4-Hydroxy-3-methylbut-trans-2-enylamino)purine (zeatin), a cytokinin isolated from Zea mays, has been synthesized by a new route. β- Methylcrotononitrile was brominated with N-bromosuccinimide yielding γ- bromo-β-methylcrotononitrile, from which trans-γ-acetoxy-β- methylcrotononitrile was prepared. The alcohol derived from this acetate was converted into trans-β-methyl-γ-(tetrahydropyran-2- yloxy)crotononitrile. Reduction and acid hydrolysis gave 4-amino-2- methylbut-trans-2-en-1-ol which was made to react with 6-chloropurine to yield zeatin. Several γ-alkoxy-β-methylcrotononitriles were prepared and reduced by aluminium chloride-lithium aluminium hydride to the corresponding unsaturated amines. Saturated nitrile formation also occurred in these reductions. The amines prepared were condensed with 6-chloropurine to yield a series of O-alkylzeatins. A number of other zeatin analogues were also synthesized. Two 6-methoxyalkylamino-purines were cleaved by sodium borohydride in the presence of iodine to 6-hydroxy- alkylaminopurines.

Abstract: The growth rates of tobacco callus tissues on media containing 10(-6) to 10 mum 6-(gamma,gamma-dimethylallylamino)purine (2iP) were measured. At concentrations of 10(-4) mum and above growth rates were exponential and dependent on cytokinin concentration. At 2iP concentrations of 10(-4) to 0.33 mum, the exponential rate was maintained for 4 to 5 doublings of fresh and dry weight. After this period a linear phase, resulting in approximately 1 doubling of weight, occurred. The growth of tissues on media containing higher than 0.33 mum 2iP was exponential for only about 15 days. At the end of this time, and well before they achieved half their final weight, they exhibited growth which was less rapid than logarithmic but more rapid than linear. Comparisons with zeatin, 6-benzylaminopurine and kinetin indicated that, although the maximum growth rates obtained with relatively high concentrations (0.1-1 mum) were similar, the naturally occurring cytokinins, 2iP and zeatin, promoted faster rates at lower concentrations (10(-3)-10(-2) mum) than did 6-benzylaminopurine and kinetin.

Abstract: Application of either zeatin or 6-benzylamino-9-(tetrahydropyran- 2-yl)purine (SD 8339) to flowers of three apple varieties resulted in the formation of fruitlets with a pronounced crown around the calyx. Several applications of these cytokinins to flowers and fruitlets of variety Cox's Orange Pippin suppressed fruit elongation, diminished the depth of the calyx depression, increased fruit density, and markedly reduced cell size. A single application of SD 8339 to flowers of the variety Jonathan increased mean individual fruit weight and promoted lobe development around the calyx of the fruit. N-(Purin-6-yl)-α-phenylglycine did not cause these effects. Application of SD 8339 to flowers of the variety Splendour promoted fruit elongation and lobe development and resulted in the development of some seedless fruit. The following stimulants of cell division applied to flowers and fruitlets of the variety Sturmer had no marked effect on cell size in the mature fruit: coconut milk, N,N'-diphenylurea, au...
