Journal ArticleDOI
Highly potent and selective inhibition of varicella-zoster virus by bicyclic furopyrimidine nucleosides bearing an aryl side chain
Christopher McGuigan,Hubert Barucki,Sally Blewett,A Carangio,Jonathan T. Erichsen,Graciela Andrei,Robert Snoeck,E. De Clercq,Jan Balzarini +8 more
TL;DR: From a range of DNA and RNA viruses tested, only VZV was inhibited by these compounds indicating their extreme selectivity for this target virus.
Journal ArticleDOI
Fluorescent bicyclic furo pyrimidine deoxynucleoside analogs as potent and selective inhibitors of VZV and potential future drugs for the treatment of chickenpox and shingles
Christopher McGuigan,Andrea Brancale,Hubert Barucki,Subashini Srinivasan,Garry Jones,Ranjith N. Pathirana,Sally Blewett,Raquel Álvarez,Christopher John Yarnold,A Carangio,Sonsoles Velázquez,Graciela Andrei,Robert Snoeck,Erik De Clercq,Jan Balzarini +14 more
Journal ArticleDOI
Lack of Susceptibility of Bicyclic Nucleoside Analogs, Highly Potent Inhibitors of Varicella-Zoster Virus, to the Catabolic Action of Thymidine Phosphorylase and Dihydropyrimidine Dehydrogenase
Jan Balzarini,Rebecca Sienaert,Sandra Liekens,André Van Kuilenburg,A Carangio,Robert Esnouf,Erik De Clercq,Christopher McGuigan +7 more
TL;DR: It is evident that BCNAs represent highly stable anti-VZV compounds that are not susceptible to breakdown by nucleoside/nucleobase catabolic enzymes and are not expected to interfere with cellular catabolic processes such as those involved in FU catabolism.
Journal ArticleDOI
Furano pyrimidines as novel potent and selective anti-VZV agents
Christopher McGuigan,Andrea Brancale,Hubert Barucki,Sheila Srinivasan,G. Jones,Ranjith N. Pathirana,A Carangio,Sally Blewett,G Luoni,O Bidet,A. Jukes,C. A. Jarvis,Graciela Andrei,Robert Snoeck,E. De Clercq,Jan Balzarini +15 more
TL;DR: The discovery, synthesis, characterization, antiviral profile, SAR, mechanism of action and development prospects for this new family of antivirals are reviewed.
Journal ArticleDOI
Synthesis and Antiviral Evaluation of Some 3′‐Fluoro Bicyclic Nucleoside Analogues
TL;DR: The synthesis of 3‐fluoro analogues of recently discovered highly potent anti‐VZV furanopyrimidine deoxynucleosides (BCNAs) is reported, strongly supporting the notion of the importance of the presence of a 3′‐OH for antiviral activity.
