Impatiens balsamina L. is a tropical ornamental and traditional medicinal herb rich in natural compounds, especially 2-methoxy-1,4-naphthoquinone (MNQ) which is a bioactive compound with tested anticancer activities. Characterization of key genes involved in the shikimate and 1,4-dihydroxy-2-naphthoate (DHNA) pathways responsible for MNQ biosynthesis and their expression profiles in I. balsamina will facilitate adoption of genetic/metabolic engineering or synthetic biology approaches to further increase production for pre-commercialization. In this study, HPLC analysis showed that MNQ was present in significantly higher quantities in the capsule pericarps throughout three developmental stages (early-, mature- and postbreaker stages) whilst its immediate precursor, 2-hydroxy-1,4-naphthoquinone (lawsone) was mainly detected in mature leaves. Transcriptomes of I. balsamina derived from leaf, flower, and three capsule developmental stages were generated, totalling 59.643 Gb of raw reads that were assembled into 94,659 unigenes (595,828 transcripts). A total of 73.96% of unigenes were functionally annotated against seven public databases and 50,786 differentially expressed genes (DEGs) were identified. Expression profiles of 20 selected genes from four major secondary metabolism pathways were studied and validated using qRT-PCR method. Majority of the DHNA pathway genes were found to be significantly upregulated in early stage capsule compared to flower and leaf, suggesting tissue-specific synthesis of MNQ. Correlation analysis identified 11 candidate unigenes related to three enzymes (NADH-quinone oxidoreductase, UDP-glycosyltransferases and S-adenosylmethionine-dependent O-methyltransferase) important in the final steps of MNQ biosynthesis based on genes expression profiles consistent with MNQ content. This study provides the first molecular insight into the dynamics of MNQ biosynthesis and accumulation across different tissues of I. balsamina and serves as a valuable resource to facilitate further manipulation to increase production of MNQ.

Comparative transcriptome analysis to identify candidate genes involved in 2-methoxy-1,4-naphthoquinone (MNQ) biosynthesis in Impatiens balsamina L

Objectives: The aim of this research work was to carry out the phytochemical screening and to evaluate the antibacterial and antifungal activity of the whole plant extract (shoot, flower, and tuber) of the flame lily (Gloriosa superba). \nMaterials and Methods: In the current work, phytochemicals were extracted from different parts of the plant using different solvents dimethyl sulfoxide, ethyl acetate, and ethanol (ETOH). These phytochemicals contained alkaloids, flavonoids, terpenoids, tannins, saponins, aromatic acids, phenolic compounds, xanthoproteins, triterpenoids, amino acids, philobatinins, carbohydrate, reducing sugar, and proteins and they were separated using standard methods. Furthermore, antimicrobial activities of methanolic separation were determined using various species of bacteria and fungi. Agar well diffusion method was used for the antimicrobial activity and the zone of the inhibition was analyzed. \nResults: The evaluation of preliminary phytochemical screening of extracts indicated the presence of carbohydrates, reducing sugars, proteins, amino acids, steroids, flavonoids, terpenoids, saponins, alkaloids, tannins, and phlobatannins. Tuber extracts highlighted effective antibacterial and antifungal activities compared with shoot and flower extracts against all the tested bacteria and fungi. ETOH extract of the tuber observed highest antibacterial activity against Staphylococcus aureus (19 mm), followed by Escherichia coli (18 mm), Micrococcus luteus (17 mm), Pseudomonas aeruginosa (17 mm), and Salmonella abony (16 mm) when compared to shoot and flower extracts. G. superba tuber extracts highlighted effective antifungal activities compared with shoot and flower extracts against all the tested fungi. ETOH extract of the tuber observed highest antifungal activity against Rhizopus oryzae (20.17 mm), followed by Mucor Sp. (19.87 mm), Aspergillus niger (18.02), Candida krusei (17.98 mm), and Candida albicans (16.88 mm) as compared to shoot and flower extracts. \nConclusion: Results of the current studies flame lily ETOH extracts showed that the plant has significant antimicrobial activities. The strong antibacterial and antifungal activities of flame lily are due to the presence of tannins and flavonoids present in.

Phytochemical analysis and evaluation of antimicrobial activity in the whole plant extracts of gloriosa superba

Rose balsam (Impatiens balsamina) is an important ornamental species grown worldwide for its attractive flowers and also having medicinal properties. Flat stem, little leaf, and phyllody symptoms were observed in I. balsamina nurseries in Uttar Pradesh and Tripura states of India during surveys from 2018 to 2020, with an incidence from 6 to 27%. Amplicons of ~ 1.2 kb were amplified in all the tested symptomatic samples of I. balsamina using universal phytoplasma primer pairs from different surveyed locations, but not from the asymptomatic plants. Pairwise sequence comparison, phylogeny, and virtual RFLP analysis of 16S rRNA gene sequences identified the phytoplasmas as 16SrI-B subgroup strain from Tripura (Lembucherra) and 16SrII-D subgroup strain from Uttar Pradesh (Gorakhpur and Faizabad). Phytoplasma presence and identity was further confirmed by amplifying secA, rp, secY, and tuf genes. This is the first report of 16SrI-B and 16SrII-D phytoplasmas detection in I. balsamina in the world.

The increasing rate of drug-repellent pathogens and poisonousness of existing antiseptic compounds has strained attention toward activity of antimicrobial products which are natural. Main purpose of this research was to assess antimicrobial activity of seeds and leaves of Coriandrum sativum’s essential oil, antioxidant, antimicrobial activity and chemical composition of Coriandrum sativum’s ethanol extracts and essential oils. Numerous approaches were used in reviewing the antioxidant activity such as, p-anisidine test – malonaldhyde, DPPH and peroxide value. Antimicrobial activity of the extracts towards six microbial strains; two bacterial strains (Salmonella typhi and Staphylococcus aureus), one yeast (Candida tropicals) and three fungal strains (Aspergillus flavus, Mucor sp and Emericella nidulans) was assessed by determination of inhibition zone and count of bacteria, yeast and spares of fungus. The antimicrobial mechanisms found in these essential oils have been explained on the basis of their content in natural compounds such as carvacrol, thymol, p-cymene and c-terpinene, among others. Although these two essential oils have received much attention, scientists working in the fields of biomedicine and food science are paying increasing attention to a wider variety of aromatic natural oils in an effort to identify original and natural applications for the inhibition of microbial pathogens. In conclusion, utilization of coriander or their components as food additives will increase the antioxidant and the antimicrobial potential of the food which prevent food deterioration and improve the shelf-life of food beside its nutritional value. The results revealed that the leaves extracts have high levels of phenolics than the seeds extract. Concerning antioxidant activity, significant decreases (p>0.001) were observed in peroxide, P- anisidine and TBA values as compared to control oil. On the other hand, scavenging activity % of the four extracts on DPPH radical were higher than that of butylated hydroxyl toluene (BHT) especially with high concentration (1000 μg/ ml). Regarding antimicrobial activity, the results showed that the extract of coriander seeds has the highest reduction percent in growth of all the examined microorganisms. The result also revealed that Mucor sp was resistant to the action of parsley extracts while Aspergillus flavus has the highest resistance against coriander extracts.

Implication of ITS phylogeny for biogeographic analysis, and comparative study of morphological and molecular interspecies diversity in Indian Impatiens

This main objective of this study is to compare the antimicrobial activity of extracts of Coriandrum sativum Linn. and Apium graveolens Linn. (family Apiaceae). Ethanol was used as organic solvent to prepare the crude extracts from the leaves. Antimicrobial activity of both plant extract were evaluated by agar well disc diffusion method against Streptococcus mutans, Klebsiella pneumonia and Pseudomonas aeroginosa. The ethanolic crude extract of Coriandrum sativum Linn. showed more activity against Streptococcus mutans and that of Apium graveolens Linn. showed more activity against Klebsiella pneumonia and Pseudomonas aeroginosa. In conclusion both these plants can be considered as natural source of antimicrobials therefore will be included in our daily diet.

I. balsamina is used in traditional methods such as Ayurveda, Unani and Siddha for various diseases and physiological conditions such as Jaundice, Corns, Snake bite etc. Phytochemical studies revealed the presence of many valuable compounds like naphthoquinones, coumarins, phenolic acids, flavonoids, anthocyanidins and steroids. The different parts of the plants like leaves, stem juice, flower are used in different places. I. balsamina have been reported to have various pharmacological activities such as antibacterial, antimicrobial, antifungal, analgesic, anti-inflammatory, antioxidant, antipruritic effects. The current review summarizes published information about the ethanopharmacology, chemical constituents, biological activities and toxicological study I.balsamina. The present review summarizes all the research work carried out on this plant in order to provide updated information for future works.
