Journal ArticleDOI
Metabolic Modulation of Clear-cell Renal Cell Carcinoma with Dichloroacetate, an Inhibitor of Pyruvate Dehydrogenase Kinase
Adam Kinnaird,Peter Dromparis,Bruno Saleme,Vikram Gurtu,Kristalee Watson,Roxane Paulin,Sotirios Zervopoulos,Trevor H. Stenson,Gopinath Sutendra,Desmond Pink,Katia Carmine-Simmen,Ronald B. Moore,John D. Lewis,Evangelos D. Michelakis +13 more
TL;DR: DCA can reverse the mitochondrial suppression of ccRCC and decrease HIF transcriptional activity, bypassing its constitutive expression and its previous clinical use in humans makes it an attractive candidate for translation toccRCC patients.
Journal ArticleDOI
FoxO1 Regulates Myocardial Glucose Oxidation Rates via Transcriptional Control of Pyruvate Dehydrogenase Kinase 4 Expression
Keshav Gopal,Bruno Saleme,Rami Al Batran,Hanin Aburasayn,Amina Eshreif,Kim L. Ho,Wayne K. Ma,Malak Almutairi,Farah Eaton,Manoj Gandhi,Edwards A. Park,Edwards A. Park,Gopinath Sutendra,John R. Ussher +13 more
TL;DR: It is demonstrated that pyruvate dehydrogenase kinase 4 is a direct transcriptional target of FoxO1 (but not FoxO3/FoxO4) in the heart, thereby controlling PDH activity and subsequent glucose oxidation rates.
Journal ArticleDOI
Tissue-specific regulation of p53 by PKM2 is redox dependent and provides a therapeutic target for anthracycline-induced cardiotoxicity.
Bruno Saleme,Vikram Gurtu,Yongneng Zhang,Adam Kinnaird,Aristeidis E. Boukouris,Keshav Gopal,John R. Ussher,Gopinath Sutendra +7 more
TL;DR: Redox-dependent differences in common proteins expressed in the myocardium and tumor can be exploited therapeutically for tissue selectivity in Chemotherapy-induced cardiotoxicity, and it is proposed that differences in oxygen tension between the mycardium and the tumor could provide a platform for redox- dependent tissue-specific therapies.
Journal ArticleDOI
Pimozide Alleviates Hyperglycemia in Diet-Induced Obesity by Inhibiting Skeletal Muscle Ketone Oxidation.
Rami Al Batran,Keshav Gopal,Megan E. Capozzi,Jadin J. Chahade,Bruno Saleme,S. Amirhossein Tabatabaei-Dakhili,Amanda A. Greenwell,Jingjing Niu,Malak Almutairi,Nikole J. Byrne,Grant Masson,Ryekjang Kim,Farah Eaton,Erin E. Mulvihill,Léa Garneau,Andrea R. Masters,Zeruesenay Desta,Carlos A. Velázquez-Martínez,Céline Aguer,Peter A. Crawford,Gopinath Sutendra,Jonathan E. Campbell,Jason R.B. Dyck,John R. Ussher +23 more
TL;DR: A fundamental contribution of enhanced ketone body oxidation to the pathology of obesity-induced T2D is defined, while suggesting pharmacological SCOT inhibition as a new class of anti-diabetes therapy is suggested.
Journal ArticleDOI
The antianginal ranolazine mitigates obesity-induced nonalcoholic fatty liver disease and increases hepatic pyruvate dehydrogenase activity
Rami Al Batran,Keshav Gopal,Hanin Aburasayn,Amina Eshreif,Malak Almutairi,Amanda A. Greenwell,Scott A. Campbell,Bruno Saleme,Emily A Court,Farah Eaton,Peter E. Light,Gopinath Sutendra,John R. Ussher +12 more
TL;DR: Ranolazine reverses obesity-induced glucose intolerance, which was associated with reduced body weight and hepatic steatosis, as well as increased hepatic pyruvate dehydrogenase (PDH) activity, which may contribute to its documented glucose-lowering actions.
