Journal ArticleDOI
Structural basis of multivalent binding to wheat germ agglutinin.
David Schwefel,Caroline Maierhofer,Johannes G. Beck,Sonja Seeberger,Kay Diederichs,Heiko M. Möller,Wolfram Welte,Valentin Wittmann +7 more
TL;DR: The findings show that bridging adjacent protein binding sites by multivalent ligands is a valid strategy to find high-affinity protein ligands and that even subtle changes of the linker structure can have a significant impact on the binding affinity.
Journal ArticleDOI
Rapid screening of lectins for multivalency effects with a glycodendrimer microarray
Núria Parera Pera,Hilbert M. Branderhorst,Raymond Kooij,Caroline Maierhofer,Marjolein van der Kaaden,Rob M. J. Liskamp,Valentin Wittmann,Rob Ruijtenbeek,Roland J. Pieters +8 more
TL;DR: To evaluate glycodendrimers as multivalent inhibitors of carbohydrate binding proteins, they were displayed on a microarray surface and a binding profile was obtained showing both the specificity of the lectin for a certain sugar and whether it prefers multivalent ligands or not.
Journal ArticleDOI
Probing multivalent carbohydrate-lectin interactions by an enzyme-linked lectin assay employing covalently immobilized carbohydrates.
TL;DR: This ELLA displayed a better differentiation between ligands of different valency leading to significantly higher relative inhibitory potencies of multivalent ligands compared to monovalent, and problems associated with the use of PSM could be overcome with this type of ELLa.
