Mitsunobu and Related Reactions: Advances and Applications

HIV/AIDS pandemics is a serious threat to health and development of mankind, and searching for effective anti-HIV agents remains actual. Considerable progress has been made in recent years in the field of drug development against HIV. A lot of structurally different coumarins were found to display potent anti-HIV activity. The current review demonstrates the variety of synthetic coumarins having unique mechanism of action referring to the different stages of HIV replication. Recent studies based on the account of various synthetic coumarins seem to indicate that some of them serve as potent non-nucleoside RT-inhibitors, another as inhibitors of HIV-integrase or HIV-protease. The merits of selecting potential anti-HIV agents to be used in rational combination drugs design and structure-activity relationships are discussed.The scientific community is looking actively for new drugs and combinations for treatment of HIV infection effective for first-line treatment, as well as against resistant mutants. The investigation on chemical anti-HIV agents gives hope and optimism about it. This review article describes recent progress in the discovery, structure modification, and structure-activity relationship studies of potent anti-HIV coumarin derivatives.

Structure-Activity Relationships of Synthetic Coumarins as HIV-1 Inhibitors

New materials for L-proline recycling have been developed. These materials have been applied to the L-proline-catalyzed aldol reaction between acetone and several aldehydes. The L-proline has been supported on the surface of modified silica gels with a monolayer of covalently attached ionic liquid with or without additional adsorbed ionic liquid. Good yields and ee values, comparable with those obtained under homogeneous conditions, have been obtained especially with imidazolinium-modified and 4-methylpyridinium-modified silica gels. Moreover, these materials have been easily recovered by simple filtration and studies about their reuse have been carried out. These studies showed that these materials can be used at least up to seven times. Finally, in the case of imidazolinium-modified silica gel, investigations showed that this support can be regenerated giving again a recyclable material with high isolated yields and reproducible ee values up to 13 cycles.

Pyranocoumarin, a novel anti-TB pharmacophore : Synthesis and biological evaluation against Mycobacterium tuberculosis

Anti-HIV natural product (+)-calanolide A is active against both drug-susceptible and drug-resistant strains of Mycobacterium tuberculosis

The synthesis of (+)-calanolide A ( 1 ), an anti-HIV-1 agent, is described. A TiCl 4 -mediated aldol reaction of compound 2 stereoselectively produced the desired syn diastereomer (±)- 5 , which was resolved by a lipase-catalyzed acylation reaction. Under Mitsunobu conditions (Ph 3 P/DEAD), the syn aldol product (+)- 5 led to the formation of trans -2,3-dimethyl chroman-4-one [(+)- 3 ] with 94% ee , while the anti aldol product (+)- 6 yielded both trans and cis derivatives (+)- 3 and (+)- 4 with 60% and 68% ee , respectively. Luche reduction on (+)- 3 led to (+)- 1 and (+)-calanolide B in a ratio of 9:1.
