Journal ArticleDOI

Human insulin from a precursor overexpressed in the methylotrophic yeast Pichia pastoris and a simple procedure for purifying the expression product.

TL;DR: In this article, the methylotrophic yeast Pichia pastoris was used to express an insulin precursor, which reached 1.5 g/L in high-density fermentation using a simple culture medium composed mainly of salt and methanol.

COMMUNICATION TO THE EDITOR Human Insulin from a Precursor Overexpressed in the Methylotrophic Yeast Pichia pastoris and a Simple Procedure for Purifying the Expression Product

TL;DR: The methylotrophic yeast Pichia pastoris, which proved successful in producing many heterologous proteins, was used to express an insulin precursor and human insulin with full receptor binding capacity and biological activity was obtained.
Journal Article

Secretory Expression of Human Insulin in Methylotrophic Yeast Pichia pastoris.

TL;DR: The porcine insulin precursor (PIP) gene and its derivative form sp-PIP gene, which had a nona-peptide (called spacer peptide, sp) added at the 5' terminus of PIP gene were inserted into the plasmid pPIC9 of Pichia pastoris to obtain secretory plasmids pPic9/PIP and pP IC9/sp-Pip, respectively.
