Journal ArticleDOI
Human insulin from a precursor overexpressed in the methylotrophic yeast Pichia pastoris and a simple procedure for purifying the expression product.
Yan Wang,Zhen-He Liang,You-Shang Zhang,Shi-Yin Yao,Ying-Gao Xu,Yue-Hua Tang,Shangquan Zhu,Da-Fu Cui,You-Min Feng +8 more
TL;DR: In this article, the methylotrophic yeast Pichia pastoris was used to express an insulin precursor, which reached 1.5 g/L in high-density fermentation using a simple culture medium composed mainly of salt and methanol.
COMMUNICATION TO THE EDITOR Human Insulin from a Precursor Overexpressed in the Methylotrophic Yeast Pichia pastoris and a Simple Procedure for Purifying the Expression Product
Yan Wang,Zhen-He Liang,You-Shang Zhang,Shi-Yin Yao,Ying-Gao Xu,Yue-Hua Tang,Shangquan Zhu,Da-Fu Cui,You-Min Feng +8 more
TL;DR: The methylotrophic yeast Pichia pastoris, which proved successful in producing many heterologous proteins, was used to express an insulin precursor and human insulin with full receptor binding capacity and biological activity was obtained.
Journal Article
Secretory expression of a single-chain insulin precursor in yeast and its conversion into human insulin
Ya-Ping Zhang,Huifeng Hu,R. Cai,You-Min Feng,Shangquan Zhu,Qing-Yu He,Yue-Hua Tang,Ming Xu,Y. Xu,Xiang Zhang,Bin Liu,Zhiqing Liang +11 more
TL;DR: A synthetic single-chain porcine insulin precursor (PIP) gene and an alpha-mating factor leader sequence (alpha MFL) gene obtained by the PCR method are inserted between the promoter and 3'-terminating sequence of the alcohol dehydrogenase gene ADH1 in plasmid pVT102-U to form plasmids pVT 102-U/alpha M FL-PIP.

Secretion expression of precursor gene of insulin in yeast and preparing process for human insulin
TL;DR: The secretion expression of single stranded insulin precursorgene in yeast and the process for preparing human insulin include gene synthesis, expression plasmid and engineering bacterium construction, and separation, purification and conversion of expression product into crystal human insulin this paper.
Journal ArticleDOI
Protein engineering of insulin: Two novel fast-acting insulins [B16Ala]insulin and [B26Ala]insulin
TL;DR: In vivo biological assay showed that the two fast-acting insulins have full or nearly full biological activity, so both [B16Ala]insulin and [B26Ala?]insulin can be well developed as fast- acting insulin for clinic use.
