Journal ArticleDOI
Spiro[pyrrolidine-2,2'-adamantanes]: synthesis, anti-influenza virus activity and conformational properties.
Ioannis Stylianakis,Antonios Kolocouris,Nicolas Kolocouris,G. Fytas,George B. Foscolos,Elizaveta Padalko,Johan Neyts,Erik De Clercq +7 more
TL;DR: This new series of anti-influenza virus A compounds is characterized by a lipophilic part, that is the pyrrolidine ring, in addition to adamantane, that can interact with influenza A M2 protein, an ultimate future goal would be the in vitro mapping of M2 Lipophilic pocket.
Journal ArticleDOI
Heterocyclic rimantadine analogues with antiviral activity
George Stamatiou,George B. Foscolos,G. Fytas,Antonios Kolocouris,Nicolas Kolocouris,Christophe Pannecouque,Myriam Witvrouw,Elizaveta Padalko,Johan Neyts,Erik De Clercq +9 more
TL;DR: It appears that changing the five-, to four- to a three-membered ring results in a drop of activity against influenza A virus.
Journal ArticleDOI
The Interferon Inducer Ampligen [Poly(I)-Poly(C12U)] Markedly Protects Mice against Coxsackie B3 Virus-Induced Myocarditis
Elizaveta Padalko,Dieter Nuyens,Armando M. De Palma,Erik Verbeken,Joeri L. Aerts,Erik De Clercq,Peter Carmeliet,Johan Neyts +7 more
TL;DR: The observed efficacies of Ampligen and poly(IC) were corroborated by the observation that the drugs also markedly reduced the virus titers in the heart, as detected by quantitative real-time reverse transcription-PCR and titration for infectious virus content.
Journal ArticleDOI
Are the 2-isomers of the drug rimantadine active anti-influenza A agents?
Grigoris Zoidis,Nicolas Kolocouris,George B. Foscolos,Antonios Kolocouris,G. Fytas,P Karayannis,Elizaveta Padalko,Johan Neyts,Erik De Clercq +8 more
TL;DR: Results showing that, although 2-adamantanamine (2-amantadine) 3 is only moderately active, some 2-rimantadines analogues are effective anti-influenza A virus agents in vitro are presented.
Journal ArticleDOI
Influence of an additional 2-amino substituent of the 1-aminoethyl pharmacophore group on the potency of rimantadine against influenza virus A.
Dimitrios Tataridis,G. Fytas,Antonios Kolocouris,Christos Fytas,Nicolas Kolocouris,George B. Foscolos,Elizaveta Padalko,Johan Neyts,Erik De Clercq +8 more
TL;DR: Interestingly, diamino compound 16 is a 4-fold more potent inhibitor than its parent monoamino heterocyclic rimantadine 4 propably because of additional hydrogen bonding interactions with the M2 protein receptor.
