Cationic Polyhexylcyanoacrylate Nanoparticles as Carriers for Antisense Oligonucleotides
TL;DR: Positive charged nanoparticles prepared from diethylaminoethyl (DEAE)-dextran and polyhexylcyanoacrylate (PHCA) were evaluated as carriers for ODNs and an active uptake mechanism, such as endocytosis, is proposed for the internalization of the ODN-nanoparticle formulations.
Journal ArticleDOI
Evaluation of aminoalkylmethacrylate nanoparticles as colloidal drug carrier systems. Part II: characterization of antisense oligonucleotides loaded copolymer nanoparticles
Hans-Peter Zobel,F Stieneker,S Atmaca-Abdel Aziz,M Gilbert,D Werner,Christian R. Noe,Joerg Kreuter,Andreas Zimmer +7 more
TL;DR: Cationic copolymer nanoparticles containing 30% (w/w) methylaminoethyl-methacrylate (MMAEMC) were found to be optimal with regard to biocompatibility and carrier properties for hydrophilic anionic antisense oligonucleotides.
Journal ArticleDOI
Enhanced antisense efficacy of oligonucleotides adsorbed to monomethylaminoethylmethacrylate methylmethacrylate copolymer nanoparticles
Hans-Peter Zobel,Monika Junghans,Vera Maienschein,D Werner,M Gilbert,Herbert Zimmermann,Christian R. Noe,Jörg Kreuter,A. Zimmer +8 more
TL;DR: Results of the cell penetration and the antisense assay demonstrated that MMAEMA nanoparticles are promising carriers for oligonucleotide administration and adsorption to nanoparticles led to an increased cellular uptake of oligon nucleotides and to a significantly enhanced antisense efficacy of unmodified phosphodiester oligon DNA as well as phosphorothioates.
Journal ArticleDOI
Evaluation of aminoalkylmethacrylate nanoparticles as colloidal drug carrier systems. Part I: synthesis of monomers, dependence of the physical properties on the polymerization methods
Hans-Peter Zobel,A. Zimmer,S Atmaca-Abdel Aziz,M Gilbert,D Werner,Christian R. Noe,Joerg Kreuter,F Stieneker +7 more
TL;DR: Positive charged copolymer nanoparticles prepared from aminoalkyl- and methylmethacrylates as well as aminoethylmethACrylamide and aminohexylmethicrylate copolymers exhibited a strong positively charged surface even at physiological pH and, therefore, are useful candidates for the adsorption of anionic drugs.
Journal ArticleDOI
Effect of ultrasonication on the stability of oligonucleotides adsorbed on nanoparticles and liposomes.
TL;DR: The oligonucleotides were protected from degradation when DEAE-stabilized PHCA-nanoparticles were employed as ODN carriers and more than 80% of the oligon nucleotides entangled in the surface matrix of these nanoparticles remained intact.
