Journal ArticleDOI
Donepezil + propargylamine + 8-hydroxyquinoline hybrids as new multifunctional metal-chelators, ChE and MAO inhibitors for the potential treatment of Alzheimer's disease.
Li Wang,Gerard Ferrer Esteban,Masaki Ojima,Oscar M. Bautista-Aguilera,Tsutomu Inokuchi,Ignacio Moraleda,Isabel Iriepa,Abdelouahid Samadi,Moussa B.H. Youdim,Alejandro Romero,Elena Soriano,Raquel Herrero,Ana Patricia Fernández,Ricardo-Martínez-Murillo,José Marco-Contelles,Mercedes Unzeta +15 more
TL;DR: DPH6 was capable to significantly decrease scopolamine-induced learning deficits in healthy adult mice and was less toxic than donepezil at high concentrations; while at low concentrations both displayed a similar cell viability profile.
Journal ArticleDOI
Design, synthesis, pharmacological evaluation, QSAR analysis, molecular modeling and ADMET of novel donepezil-indolyl hybrids as multipotent cholinesterase/monoamine oxidase inhibitors for the potential treatment of Alzheimer's disease
Oscar M. Bautista-Aguilera,Gerard Ferrer Esteban,Irene Bolea,Katarina Nikolic,Danica Agbaba,Ignacio Moraleda,Isabel Iriepa,Abdelouahid Samadi,Elena Soriano,Mercedes Unzeta,José Marco-Contelles +10 more
TL;DR: It is found that, in general, and for the same substituent, amines are more potent ChE inhibitors or equipotent than the corresponding amides, showing a clear EeAChE inhibition selectivity.
Journal ArticleDOI
Nitric oxide blocks hKv1.5 channels by S-nitrosylation and by a cyclic GMP-dependent mechanism
Lucía Núñez,Miguel Vaquero,Ricardo Gómez,Ricardo Caballero,Petra J. Mateos-Cáceres,Carlos Macaya,Isabel Iriepa,Enrique Gálvez,Antonio J. López-Farré,Juan Tamargo,Eva Delpón +10 more
TL;DR: Whether nitric oxide modulates the human potassium channel hKv1.5, which generates the ultrarapid delayed rectifier current (IKur) that determines the height and duration of atrial action potentials is analyzed.
Journal ArticleDOI
Multitarget‐Directed Ligands Combining Cholinesterase and Monoamine Oxidase Inhibition with Histamine H3R Antagonism for Neurodegenerative Diseases
Oscar M. Bautista-Aguilera,Stefanie Hagenow,Alejandra Palomino-Antolín,Víctor Farré-Alins,Lhassane Ismaili,Pierre‐Louis Joffrin,María Luisa Jimeno,Ondřej Soukup,Jana Janockova,Lena Kalinowsky,Ewgenij Proschak,Isabel Iriepa,Ignacio Moraleda,Johannes Stephan Schwed,Alejandro Romero Martínez,Francisco López-Muñoz,Mourad Chioua,Javier Egea,Rona R. Ramsay,José Marco-Contelles,Holger Stark +20 more
TL;DR: Contilisant (at 1 mg kg-1 i.p.) also significantly improved lipopolysaccharide-induced cognitive deficits and showed antioxidative neuroprotective effects as well as the ability to penetrate the blood-brain barrier.
Journal ArticleDOI
Tackling neuroinflammation and cholinergic deficit in Alzheimer's disease: Multi-target inhibitors of cholinesterases, cyclooxygenase-2 and 15-lipoxygenase.
Ehab D. AlFadly,Perihan A. Elzahhar,Anna Tramarin,Salwa Elkazaz,Hossam A. Shaltout,Marwa M. Abu-Serie,Jana Janockova,Ondrej Soukup,Doaa A. Ghareeb,Ahmed F. El-Yazbi,Rim W. Rafeh,Nour-Mounira Z. Bakkar,Firas Kobeissy,Isabel Iriepa,Ignacio Moraleda,Manal N. S. Saudi,Manuela Bartolini,Ahmed S.F. Belal +17 more
TL;DR: To the best of the authors' knowledge, compounds 5b, 5d and 6e are the first balanced, safe and multi-target compounds hitting the disease at the three mentioned hubs.
