Journal ArticleDOI
Breast and ovarian cancer risks due to inherited mutations in BRCA1 and BRCA2.
TL;DR: Physical exercise and lack of obesity in adolescence were associated with significantly delayed breast cancer onset, and risks appear to be increasing with time.
Journal ArticleDOI
Spectrum of mutations in BRCA1, BRCA2, CHEK2, and TP53 in families at high risk of breast cancer.
Tom Walsh,Silvia Casadei,Kathryn Hale Coats,Elizabeth M. Swisher,Sunday M. Stray,Jake Higgins,Kevin C. Roach,Jessica B. Mandell,Ming K. Lee,Sona Ciernikova,Lenka Foretova,Pavel Soucek,Mary Claire King +12 more
TL;DR: Among patients with breast cancer and severe family histories of cancer who test negative (wild type) for BRCA1 and BRCa2, approximately 12% can be expected to carry a large genomic deletion or duplication in one of these genes, and approximately 5% could carry a mutation in CHEK2 or TP53.
Journal ArticleDOI
Detection of inherited mutations for breast and ovarian cancer using genomic capture and massively parallel sequencing
Tom Walsh,Ming K. Lee,Silvia Casadei,Anne M. Thornton,Sunday M. Stray,C. Pennil,Alexander Nord,Jessica B. Mandell,Elizabeth M. Swisher,Mary Claire King +9 more
TL;DR: A genomic assay to capture, sequence, and detect all mutations in 21 genes, including BRCA1 and BRCa2, with inherited mutations that predispose to breast or ovarian cancer, which enables widespread genetic testing and personalized risk assessment for breast and ovarian cancer.
Journal ArticleDOI
Population-based screening for breast and ovarian cancer risk due to BRCA1 and BRCA2
Efrat Gabai-Kapara,Amnon Lahad,Bella Kaufman,Eitan Friedman,Shlomo Segev,Paul Renbaum,Rachel Beeri,Moran Gal,Julia Grinshpun-Cohen,Karen Djemal,Jessica B. Mandell,Ming K. Lee,Uziel Beller,Raphael Catane,Mary Claire King,Ephrat Levy-Lahad,Ephrat Levy-Lahad +16 more
TL;DR: It is determined that risks of breast and ovarian cancer for BRCA1 and BRCa2 mutation carriers ascertained from the general population are as high as for mutation carrier ascertained through personal or family history of cancer.
Journal ArticleDOI
Contribution of Inherited Mutations in the BRCA2-Interacting Protein PALB2 to Familial Breast Cancer
Silvia Casadei,Barbara M. Norquist,Tom Walsh,Sunday M. Stray,Jessica B. Mandell,Ming K. Lee,John A. Stamatoyannopoulos,Mary Claire King +7 more
TL;DR: Given this mutation prevalence and risk, consideration might be given to clinical testing of PALB2 by complete genomic sequencing for familial breast cancer patients with wild-type sequences at BRCA1 and BRCa2.
