Abstract: We present a novel approach for generating targeted deletions of genomic segments in human and other eukaryotic cells using engineered zinc finger nucleases (ZFNs). We found that ZFNs designed to target two different sites in a human chromosome could introduce two concurrent DNA double-strand breaks (DSBs) in the chromosome and give rise to targeted deletions of the genomic segment between the two sites. Using this method in human cells, we were able to delete predetermined genomic DNA segments in the range of several-hundred base pairs (bp) to 15 mega-bp at frequencies of 10−3 to 10−1. These high frequencies allowed us to isolate clonal populations of cells, in which the target chromosomal segments were deleted, by limiting dilution. Sequence analysis revealed that many of the deletion junctions contained small insertions or deletions and microhomologies, indicative of DNA repair via nonhomologous end-joining. Unlike other genome engineering tools such as recombinases and meganucleases, ZFNs do not require preinsertion of target sites into the genome and allow precise manipulation of endogenous genomic scripts in animal and plant cells. Thus, ZFN-induced genomic deletions should be broadly useful as a novel method in biomedical research, biotechnology, and gene therapy.

Abstract: Various scientific computations have become so complex, and thus computation tools play an important role. In this paper, we explore the state-of-the-art framework providing high-level matrix computation primitives with MapReduce through the case study approach, and demonstrate these primitives with different computation engines to show the performance and scalability. We believe the opportunity for using MapReduce in scientific computation is even more promising than the success to date in the parallel systems literature.

Abstract: NAND flash-based storage devices have rapidly improved their position in the secondary storage market ranging from mobile embedded systems to personal computer and enterprise storage systems. Recently, the most important issue of NAND flash-based storage systems is the performance of random writes as well as sequential writes, which strongly depends on their two main software layers: a Buffer Management Layer (BML) and a Flash Translation Layer (FTL). The primary goal of our study is to highly improve the overall performance of NAND flash-based storage systems by exploiting the cooperation between those two layers. In this paper, we propose an FTL-aware BML policy called Selective Block Padding and a BML-based FTL algorithm called Optimized Switch Merge, which overcome the limitations of existing approaches on performance enhancement. When using both the proposed techniques, evaluation results show that the throughput is significantly increased over that of previous studies.

Abstract: Objectives : Thus this study attempted to look into the level of community residents' dental health and the state of their dental diseases and estimate the general dental health condition so that these data can be available as references in upcoming public dental health planning, and to provide fundamental data for promotion of the level of residents' dental health by performing dental health education. Methods : Among community residents who visited the department of Dental Hygiene of Shinsung University for scaling from March to May 2009, the data of total 346 subjects of 129 women and 217 men were selected, and DMFT rate, DMFT index, dental health capacity of the first permanent molar, and tooth morality rate were investigated. For statistical analysis, SPSS 14.0 was used, for general characteristics of the subjects, frequency analysis was conducted, for caries experience in the permanent teeth of the subjects, mean and standard deviation were calculated, for caries experience in the permanent teeth by sex and residence, independent t-test was performed, and for caries experience in the permanent teeth by age, one-way ANOVA was conducted. The significance level applied to these analyses was 0.05. Results : As a result of investigation and analysis on caries in the permanent teeth of community residents who visited the department for scaling in this study, the findings are as follows: 1. For sex, DMFT rate and tooth morality rate were found to be higher in women, while dental health capacity of the first permanent molar was found to be higher in men. 2. For age, DMFT rate and DMFT index were found to be highest in 40~49 years old, while DMFT rate, DMFT index, and tooth morality rate were found to be lowest in under 20 years old. Dental health capacity of the first permanent molar was found to be highest in under 20 years old, 20-29 years old, and 30-39 years old and found to be lowest in more than their sixties as 39.78 points(p

Abstract: Purpose: In the case of repair for far distal parts of FDP (Flexor digitorum profundus) division, the method of either pull-out suture or fixation of tendon to the distal phalanx is preferred. In this paper, the results of a modified loop suture technique used for the complete division of FDP from both zone 1a and distal parts of zone 1b in Moiemen classification are presented. Methods: From July 2006 to July 2009, the modified loop suture technique was used for the 10 cases of FDP in complete division from zone 1a and distal parts of zone 1b, especially where insertion sites were less than 1 cm apart from a tendon of a stump. In a suture technique, a loop is applied to each distal and proximal parts of tendon respectively. Core suture of 2-strand and epitendinous suture are done with PDS 4-0. Out of 10 patients, the study was done on 6 patients who were available for the followup. The average age of the patients was 49.1 years (in the range from 26 to 67). 5 males and 1 female patients were involved in this study. There were 3 cases with zone 1a and distal parts of zone 1b. The average distance to the distal tendon end was 0.6 cm. There were 5 cases underwent microsurgical repair where both artery and nerve divided. One case of only tendon displacement was presented. The dorsal protective splint was kept for 5 weeks on average. The results of the following tests were measured: active & passive range of motion, grip strength test, key pinch and pulp pinch test. Results: The follow-up period on average was 11 months, in the range from 2 to 20 months. There was no case of re-rupture, but tenolysis was performed in 1 cases. In all 6 cases, the average active range of motion of distal interphalangeal joint was 50.8 degree. The grip strength (ipsilateral/contralateral) was measured as 88.7% and the pulp pinch test was 79.2% as those of contralateral side. Flexion contracture was presented in 2 cases (15 degree on average) and there was no quadrigia effect found. Conclusion: Despite short length of tendon from the insertion site in FDS rupture in zone 1a and distal parts of zone 1b, sufficient functional recovery could be expected with the tendon to tendon repair using the modified loop suture technique.
