TDP-43 subcellular mislocalisation is correlated with loss of optineurin binding for frontotemporal dementia and amyotrophic lateral sclerosis associated TBK1 missense variants

TL;DR: Results suggest that loss of the direct interaction between TBK1 and OPTN is more critical to FTD and ALS pathogenesis than TBK2’s kinase activity, and further studies are needed to elucidate exactly how loss ofTBK1 binding to OPTN leads to TDP-43 pathology and ultimately neurodegeneration.

The major TMEM106B dementia risk allele affects TMEM106B protein levels and myelin lipid homeostasis in the ageing human hippocampus

TL;DR: In this paper , the authors investigated the effect of major dementia risk alleles in the TMEM106B and APOE genes on the hippocampal proteome and lipidome, adjusting for age, gender and post-mortem interval.
