Effects of Single-Dose Interleukin-12 Exposure on Interleukin-12–Associated Toxicity and Interferon-γ Production
John P. Leonard,Matthew L. Sherman,Gerald L. Fisher,Lynn J. Buchanan,Glenn R. Larsen,Michael B. Atkins,Jeffrey A. Sosman,Janice P. Dutcher,Nicholas J. Vogelzang,John L. Ryan +9 more
TL;DR: It is determined that a single injection of rhIL-12 2 weeks before consecutive dosing included in the Phase 1 study, but not in the schedule of administration in thephase 2 study, has a profound abrogating effect on IL-12–induced interferon-γ (IFN-γ) production and toxicity.
Journal ArticleDOI
The Antiparasitic Moxidectin: Safety, Tolerability, and Pharmacokinetics in Humans
Monette M. Cotreau,Sarah Warren,John L. Ryan,Lawrence Fleckenstein,Sreenivasa R. Vanapalli,Kenneth Brown,David Rock,Chieh-Yu Chen,Ullrich S. Schwertschlag +8 more
TL;DR: The results from this study indicate that MOX is safe and well tolerated in humans between the doses of 3 mg and 36 mg.
Journal ArticleDOI
Regulation of gene expression in mouse macrophages stimulated with bacterial CpG-DNA and lipopolysaccharide.
Jian Jun Gao,Veronica Diesl,Tatiana Wittmann,David C. Morrison,John L. Ryan,Stefanie N. Vogel,Maximillian T. Follettie +6 more
TL;DR: It is concluded that CpG‐DNA signaling through TLR9 activates a subset of genes induced by LPS‐TLR4 signaling, which triggers innate and acquired immune responses through activated expression of various genes in immune cells, including macrophages.
Journal ArticleDOI
A phase II open-label study of recombinant human interleukin-12 in patients with stage IA, IB, or IIA mycosis fungoides.
Madeleine Duvic,Matthew L. Sherman,Gary S. Wood,Timothy M. Kuzel,Elise A. Olsen,Francine M. Foss,Robert Laliberte,John L. Ryan,Kristilyn Zonno,Alain H. Rook +9 more
TL;DR: In this article, the safety and efficacy of subcutaneous recombinant human IL-12 (rhIL-12) in early mycosis fungoides (MF; stage IA-IIA) in a multi-center, open label clinical trial were evaluated.
Journal ArticleDOI
Bacterial LPS and CpG DNA differentially induce gene expression profiles in mouse macrophages.
Jian Jun Gao,Veronica Diesl,Tatiana Wittmann,David C. Morrison,John L. Ryan,Stefanie N. Vogel,Maximillian T. Follettie +6 more
TL;DR: Macrophage gene expression profiles studied indicate that the CpG DNA signaling pathway through TLR9 activates only a subset of genes induced by the LPS TLR4 signaling pathway.
