Journal ArticleDOI
The Human Skin Barrier Is Organized as Stacked Bilayers of Fully Extended Ceramides with Cholesterol Molecules Associated with the Ceramide Sphingoid Moiety
Ichiro Iwai,HongMei Han,Lianne den Hollander,Stina Svensson,Stina Svensson,Lars-Goran Ofverstedt,Jamshed Anwar,Jonathan R. Brewer,Maria Bloksgaard,Aurelie Laloeuf,Daniel Nosek,Daniel Nosek,Sergej Masich,Luis A. Bagatolli,Ulf Skoglund,Lars Norlén,Lars Norlén +16 more
TL;DR: The determination of the molecular organization of the skin's lipid matrix in situ, in its near-native state, is reported on, using a methodological approach combining very high magnification cryo-electron microscopy of vitreous skin section defocus series, molecular modeling, and EM simulation.
Journal ArticleDOI
Insights into the Cellular Response Triggered by Silver Nanoparticles using Quantitative Proteomics
Thiago Verano-Braga,Rona Miethling-Graff,Katarzyna Wojdyla,Adelina Rogowska-Wrzesinska,Jonathan R. Brewer,Helmut Erdmann,Frank Kjeldsen +6 more
TL;DR: It is reported that proteins involved in SUMOylation were up-regulated after exposure to 20 nm silver nanoparticles, and some unique cellular processes are driven by the size.
Journal ArticleDOI
Exposure to silver nanoparticles induces size- and dose-dependent oxidative stress and cytotoxicity in human colon carcinoma cells
Rona Miethling-Graff,Rita Rumpker,Madeleine Richter,Thiago Verano-Braga,Frank Kjeldsen,Jonathan R. Brewer,James Hoyland,Horst-Günter Rubahn,Helmut Erdmann +8 more
TL;DR: It was hypothesized that this size-dependent effect resulted from the stimulation of a signaling cascade that generated ROS and inflammatory markers, leading to mitochondrial dysfunction and subsequently inducing apoptosis, and this data indicated that cellular uptake and toxicity were regulated by size.
Journal ArticleDOI
Plasticity of Epididymal Adipose Tissue in Response to Diet-Induced Obesity at Single-Nucleus Resolution
Anitta Kinga Sárvári,Elvira Laila Van Hauwaert,Lasse K. Markussen,Ellen Gammelmark,Ann-Britt Marcher,Morten F. Ebbesen,Ronni Nielsen,Jonathan R. Brewer,Jesper Grud Skat Madsen,Susanne Mandrup +9 more
TL;DR: In this paper, a single-nucleus RNA-seq was applied to map the plasticity of mouse epididymal white adipose tissue at single nucleus resolution in response to highfat-diet-induced obesity.
Journal ArticleDOI
Escherichia coli Uropathogenesis In Vitro: Invasion, Cellular Escape, and Secondary Infection Analyzed in a Human Bladder Cell Infection Model
Thomas Emil Andersen,Surabhi Khandige,Michelle Madelung,Jonathan R. Brewer,Hans Jørn Kolmos,Jakob Møller-Jensen +5 more
TL;DR: Under growth conditions established to resemble those present in vivo, the elements of the proposed uropathogenic cascade were inducible in a human BEC model system and shown how these characteristics are reproduced in vitro.
