Journal ArticleDOI
GM-CSF inhibition reduces cytokine release syndrome and neuroinflammation but enhances CAR-T cell function in xenografts
Rosalie M. Sterner,Reona Sakemura,Michelle J. Cox,Nan Yang,Roman H. Khadka,Cynthia L. Forsman,Michael J. Hansen,Fang Jin,Katayoun Ayasoufi,Mehrdad Hefazi,Kendall J. Schick,Denise K. Walters,Omar H. Ahmed,Dale Chappell,Tarek Sahmoud,Cameron Durrant,Wendy K. Nevala,Mrinal M. Patnaik,Larry R. Pease,Karen E. Hedin,Neil E. Kay,Aaron J. Johnson,Saad S. Kenderian +22 more
TL;DR: It is shown that GM-CSF neutralization with lenzilumab does not inhibit CART19 cell function in vitro or in vivo, and a novel approach to abrogate NI and CRS through GM- CSF neutralized, which may potentially enhance CAR-T cell function.
Journal ArticleDOI
Targeting Cancer-Associated Fibroblasts in the Bone Marrow Prevents Resistance to CART-Cell Therapy in Multiple Myeloma.
Reona Sakemura,Mehrdad Hefazi,Elizabeth L. Siegler,Michelle J. Cox,Daniel P. Larson,Michael J. Hansen,Claudia Manriquez Roman,Kendall J. Schick,Ismail Can,Erin E. Tapper,Paulina Horvei,Mohamad Adada,Evandro D. Bezerra,Lionel A. Kankeu Fonkoua,Michael W. Ruff,Wendy K. Nevala,Denise K. Walters,Sameer A. Parikh,Yi Lin,Diane F. Jelinek,Neil E. Kay,P. Leif Bergsagel,Saad S. Kenderian +22 more
TL;DR: It is demonstrated for the first time that dual targeting both malignant plasma cells and the CAFs within the TME is a novel strategy to overcome resistance to CART-cell therapy in MM.
Journal ArticleDOI
Leukemic extracellular vesicles induce chimeric antigen receptor T cell dysfunction in chronic lymphocytic leukemia.
Michelle J. Cox,Michelle J. Cox,Fabrice Lucien,Reona Sakemura,Justin C. Boysen,Yohan Kim,Paulina Horvei,Claudia Manriquez Roman,Michael J. Hansen,Erin E. Tapper,Elizabeth L. Siegler,Cynthia L. Forsman,Sydney B. Crotts,Kendall J. Schick,Mehrdad Hefazi,Michael W. Ruff,Ismail Can,Mohamad M. Adada,Evandro D. Bezerra,Lionel A. Kankeu Fonkoua,Wendy K. Nevala,Esteban Braggio,Wei Ding,Sameer A. Parikh,Neil E. Kay,Saad S. Kenderian +25 more
TL;DR: In this article, the authors used chronic lymphocytic leukemia (CLL) as a model to investigate the interactions between the tumor microenvironment and CART cells and found that CLL-derived EVs were able to induce a state of CART cell dysfunction characterized by phenotypical, functional and transcriptional changes of exhaustion.
Journal ArticleDOI
Targeting Cancer Associated Fibroblasts in the Bone Marrow Prevents Resistance to Chimeric Antigen Receptor T Cell Therapy in Multiple Myeloma
Reona Sakemura,Michelle J. Cox,Michael J. Hansen,Mehrdad Hefazi,Claudia Manriquez Roman,Kendall J. Schick,Erin E. Tapper,Paulina Roman Moreno,Michael W. Ruff,Denise K. Walters,Sameer A. Parikh,Neil E. Kay,Saad S. Kenderian +12 more
TL;DR: CART cell therapy targeting both malignant MM cells and their CAFs is aimed to develop and whether this strategy can reverse MM-CAF induced CART cell inhibition is investigated.
Journal ArticleDOI
Development of a Clinically Relevant Reporter for Chimeric Antigen Receptor T-cell Expansion, Trafficking, and Toxicity.
Reona Sakemura,Aditya Bansal,Elizabeth L. Siegler,Mehrdad Hefazi,Nan Yang,Nan Yang,Roman H. Khadka,Alysha N Newsom,Michael J. Hansen,Michelle J. Cox,Claudia Manriquez Roman,Kendall J. Pandey,Timothy R. DeGrado,Lukkana Suksanpaisan,Neil E. Kay,Kah-Whye Peng,Stephen J. Russell,Saad S. Kenderian +28 more
TL;DR: NIS provides a sensitive, clinically applicable platform for CART-cell imaging with PET scan and confirms NIS+BCMA-Cart-cell trafficking to the tumor sites in localized and systemic tumor models.
