PCR-SSCP and Sequencing Analysis For Studying Leptin Gene Polymorphism and Its Association with Reproductive Status of Egyptian Buffalo

Objective: This study was aimed to identify IGF-1, LEP and PRLR polymorphisms by PCR-RFLP method in Saanen goats.Material and Methods: In the study, nasal samples were collected from 72 Saanen goats via swabs and DNA was extracted. Polymorphisms of IGF-1, LEP and PRLR were determined by PCR amplification followed by RFLP method using restriction enzymes Cac8I, NmuCI and Hin1II, respectively. Results: LEP/NmuCI produced three genotypes as TT, TC and CC with frequencies 90.3, 6.9 and 2.8%, respectively and two alleles (T=93.8%, C=6.2%). The frequencies of genotype for PRLR gene were CC (90.3%), CT (5.5%) and TT (4.2%) and allele frequencies were C (93.1%) and T (6.9%). The IGF-1/Cac8I locus was monomorphic in Saanen goat population. Conclusion: The present results may be useful for future studies conducted on goat genome with respect to IGF-1, LEP and PRLR genes.

IGF-1/CaC8I, LEP/NmuCI and PRLR/Hin1II Polymorphisms in Saanen Goats

This study was designed to identify single-nucleotide polymorphisms (SNPs) of some candidate genes related to lipid metabolism and their association with carcass fat in male crossbred lambs. Hence, 96 of almost 11-month-old Booroola Merino-Afshari crossbred lambs (first-generation backcross) were used by considering their phenotypic carcass traits. Then, DNA was extracted and DNA targets were amplified using designed specific primers by PCR procedure. Identification of potential SNPs was done by a direct sequencing method for LEP, FABP4, DGAT1, GH, and TRIB3 genes using the sequencing-RFLP procedure. Then, the most probable statistical models based on additive and genotypic effects of identified SNPs in each trait were obtained by the Bayesian model averaging (BMA) approach of R software (Ver. 3.3.1) to assess the association of SNPs with traits. Detected SNPs in this study included two SNPs in exon 3 of LEP, one SNP in exon 2 of TRIB3, one SNP in intron 2 of FABP4, one SNP in 5ʹ UTR of DGAT1, and two SNPs in 3ʹ UTR of GH genes. For carcass weight trait, one of the identified SNP genotypes in the LEP (c.587G > A) had a higher probability in the model. Carcass weight of lambs with GA genotype was 2.46 kg heavier than GG genotype. Also, two genes of TRIB3 and GH2 had the highest probability in the models of fat tail and waste weight, respectively. Based on the results, these polymorphisms can be used in the marker-assisted selection of breeding programs and designing DNA chips for genomic selection.

Effect of Leptin Gene Polymorphism on Reproductive Efficiency in Awassi Ewes

This study prepared to explore the relation between Growth Differentiation Factor 9 (GDF9) genotypes and calving rate, Follicle-stimulating hormone (FSH) and Estradiol (E2) in Iraqi Holstein Friesians breed. Fifteen blood samples collected from a mother of DZTB (with high calving rate records), another blood sample collected from 15 SB cows, the DNA extracted and six primers designed for PCR and sequencing analysis. The FSH and E2 level tested through estrus phase for the two groups (10 n for each group). The sequence evaluation revealed that presence 2 single nucleotide polymorphisms (SNPs) in exon II; A (1109) T and G (1133) A. The genotypic frequency for mutant genotypes was higher significantly (P<0.01) in DZTB cows (with calving rate) than wild at the same loci. On the other hand, the wild genotypes recorded higher significant increment (P<0.01) for SB cows when compared with mutant genotypes in the same loci. Moreover, a significant rise (P<0.05) was reported in E2 and FSH levels for DZTB cows and mutant genotypes (P<0.01) against SB cows and wild genotypes respectively in 0 and 24 h of estrus phase. Non-significant differences recorded in E2 concentration between the same genotypes at same period. In conclusion, the GDF9 exon II SNPs increase the calving rate in Holstein Friesians cows. The blood FSH and E2 concentrations are higher in the mother of DZTB may control the superovulation. Finally, these SNPs can be considered as markers to accelerate the breeding programs and used in embryo transfer and in vitro embryo production for Iraqi Holstein Friesians cows breed.

Inspection the Linkage between Bovine GDF9 SNPs and Calving Rate (Superovulation) in Holstein Friesians Cows

The objective of the current study was to display the effects of addition zinc & selenium on ram semen characteristic in vitro. This study was directed on 3 local breed rams, aged between 2-4 years, presented in the farm of college of veterinary medicine / Al-Fallujah University,at a period of time from the beginning of February to march 2018. The rams were feed with alfalfa and hay.
All the experimental animal were treated from internal parasite. semen was collected from each ram by electro-ejaculator apparatus at morning hours at weekly interval.Over a course of 8 week (eight samples per ram) semen sample were put in a water bath with 38c0, pooled semen of both rams were evaluated.
The samples of Semen were diluted 1: 10 with a tris based extender according to the concentration of spermatozoa ( Tris= 24.2 gm, citric acid = 13.4 gm, fructose = 10 gm, glycerin= 64 ml, egg yolk= 192 ml, distal water up to one liter),diluted semen were taken and divided into 3 parts ( each part 2 ml), the first part (T1) added to it 4.0 µ selenium /ml  of diluent. The second part (T2) added to it 0.576µ zinc sulphate / ml of diluted semen . the third part (T3) not added to it anything and serve as a control, another fresh samples were taken and diluted and then cooled gradually via addition a piece of ice till it reaches 4co within 2 hours,semen parameters were measured after dilution and cooling.The result presenteda significant differences at (P<0.05) in an individual motility%, Dead%,  alive% & abnormalities% between Zn treated group as comparing with selenium or control group. It was concluded that adding of zinc sulphate to the diluted and cooled  semen have a beneficial effect  on someram semencharacteristic in non-breeding season.
