Conceptual density functional theory.

Over the last 20 years, the number of publications outlining the advances in design strategies, growing techniques, and characterization of cocrystals has continued to increase significantly within the crystal engineering field. However, only within the last decade have cocrystals found their place in pharmaceuticals, primarily due to their ability to alter physicochemical properties without compromising the structural integrity of the active pharmaceutical ingredient (API) and thus, possibly, the bioactivity. This review article will highlight and discuss the advances made over the last 10 years pertaining to physical and chemical property improvements through pharmaceutical cocrystalline materials and, hopefully, draw closer the fields of crystal engineering and pharmaceutical sciences.

Pharmaceutical Cocrystals and Their Physicochemical Properties

Drugs with low water solubility are predisposed to low and variable oral bioavailability and, therefore, to variability in clinical response. Despite significant efforts to "design in" acceptable developability properties (including aqueous solubility) during lead optimization, approximately 40% of currently marketed compounds and most current drug development candidates remain poorly water-soluble. The fact that so many drug candidates of this type are advanced into development and clinical assessment is testament to an increasingly sophisticated understanding of the approaches that can be taken to promote apparent solubility in the gastrointestinal tract and to support drug exposure after oral administration. Here we provide a detailed commentary on the major challenges to the progression of a poorly water-soluble lead or development candidate and review the approaches and strategies that can be taken to facilitate compound progression. In particular, we address the fundamental principles that underpin the use of strategies, including pH adjustment and salt-form selection, polymorphs, cocrystals, cosolvents, surfactants, cyclodextrins, particle size reduction, amorphous solid dispersions, and lipid-based formulations. In each case, the theoretical basis for utility is described along with a detailed review of recent advances in the field. The article provides an integrated and contemporary discussion of current approaches to solubility and dissolution enhancement but has been deliberately structured as a series of stand-alone sections to allow also directed access to a specific technology (e.g., solid dispersions, lipid-based formulations, or salt forms) where required.

Strategies to Address Low Drug Solubility in Discovery and Development

Solubility, the phenomenon of dissolution of solute in solvent to give a homogenous system, is one of the important parameters to achieve desired concentration of drug in systemic circulation for desired (anticipated) pharmacological response. Low aqueous solubility is the major problem encountered with formulation development of new chemical entities as well as for the generic development. More than 40% NCEs (new chemical entities) developed in pharmaceutical industry are practically insoluble in water. Solubility is a major challenge for formulation scientist. Any drug to be absorbed must be present in the form of solution at the site of absorption. Various techniques are used for the enhancement of the solubility of poorly soluble drugs which include physical and chemical modifications of drug and other methods like particle size reduction, crystal engineering, salt formation, solid dispersion, use of surfactant, complexation, and so forth. Selection of solubility improving method depends on drug property, site of absorption, and required dosage form characteristics.

Drug Solubility: Importance and Enhancement Techniques

How do molecules aggregate in solution, and how do these aggregates consolidate themselves in crystals? What is the relationship between the structure of a molecule and the structure of the crystal it forms? Why do some molecules adopt more than one crystal structure? Why do some crystal structures contain solvent? How does one design a crystal structure with a specified topology of molecules, or a specified coordination of molecules and/or ions, or with a specified property? What are the relationships between crystal structures and properties for molecular crystals? These are some of the questions that are being addressed today by the crystal engineering community, a group that draws from the larger communities of organic, inorganic, and physical chemists, crystallographers, and solid state scientists. This Perspective provides a brief historical introduction to crystal engineering itself and an assessment of the importance and utility of the supramolecular synthon, which is one of the most important concepts in the practical use and implementation of crystal design. It also provides a look to the future from the viewpoint of the author, and indicates some directions in which this field might be moving.

Crystal engineering: from molecule to crystal.

A crystal engineering strategy for designing cocrystals of pharmaceuticals is presented. The strategy increases the probability of discovering useful cocrystals and decreases the number of experiments that are needed by selecting API:guest combinations that have the greatest potential of forming energetically and structurally robust interactions. Our approach involves multicomponent cocrystallization of hydrochloride salts, wherein strong hydrogen bond donors are introduced to interact with chloride ions that are underutilized as hydrogen bond acceptors. The strategy is particularly effective in producing cocrystals of amine hydrochlorides with neutral organic acid guests. As an example of the approach, we report the discovery of three cocrystals containing fluoxetine hydrochloride (1), which is the active ingredient in the popular antidepressant Prozac. A 1:1 cocrystal was prepared with 1 and benzoic acid (2), while succinic acid and fumaric acid were each cocrystallized with 1 to provide 2:1 cocrystals of fluoxetine hydrochloride:succinic acid (3) and fluoxetine hydrochloride:fumaric acid (4). The presence of a guest molecule along with fluoxetine hydrochloride in the same crystal structure results in a solid phase with altered physical properties when compared to the known crystalline form of fluoxetine hydrochloride. On the basis of intrinsic dissolution rate experiments, cocrystals 2 and 4 dissolve more slowly than 1, and 3 dissolves more quickly than 1. Powder dissolution experiments demonstrated that the solid present at equilibrium corresponds to the cocrystal for 2 and 4, while 3 completely converted to 1 upon prolonged slurry in water.

Crystal Engineering Approach To Forming Cocrystals of Amine Hydrochlorides with Organic Acids. Molecular Complexes of Fluoxetine Hydrochloride with Benzoic, Succinic, and Fumaric Acids

The Site of Protonation of Aniline

Ab initio molecular orbital calculations at the MP2/6–31G*//6–31G* + ZPVE level of theory suggest that the ion (CH3)2S+CH2˙ is 75 kJ mol−1 higher in energy than ionized ethyl methyl sulfide. However, this distonic ion is stable toward isomerization to the conventional structure: the two ions show distinctly different reactivity in a dual-cell Fourier transform ion cyclotron resonance mass spectrometer. Molecular orbital calculations further indicate that the charge site of the ion is on the sulfur atom whereas the odd-spin density is localized on the CH2 group. The chemical properties of the ion reflect this description. The coordinatively saturated sulfonium charge site is chemically inert. This radical cation possesses unique chemical reactivity in that its reactions are associated with the radical site. Most of these reactions involve a homolytic bond cleavage in the neutral molecule, resulting in abstraction of an atom or a radical by the ion. The reaction efficiency correlates with the homolytic bond dissociation energy of the bond being broken in the neutral molecule, suggesting a simple radical abstraction mechanism for these reactions. Hence, the methylenedimethylsulfonium ion is best described as on electrophilic radical with an inert charge site.

Radical‐type reactivity of the methylenedimethylsulfonium ion, (CH3)2S+ ? CH2˙

The homolytic Se−H bond energy of benzeneselenol, a very efficient radical trap in solution and in the gas phase, was determined experimentally by using two independent approaches in a Fourier-transform ion cyclotron resonance mass spectrometer. The hydrogen−selenium bond strength was concluded to be 76−80 kcal mol-1 based on the measurement of the efficiency of hydrogen atom abstraction from benzeneselenol by several radical cations. The Se−H bond energy was also determined indirectly through the use of a thermochemical cycle. This approach required the measurement of the adiabatic ionization energy of benzeneselenol and the proton affinity of C6H5Se•. An adiabatic ionization energy of 8.3 ± 0.1 eV was obtained by measurement of the efficiencies of various electron transfer reactions in the forward and reverse directions. The proton affinity of C6H5Se• (acidity of the benzeneselenol radical cation) was found to be 200 ± 3 kcal mol-1 based on the measured efficiencies of several proton transfer reactions....
