Journal ArticleDOI
The Dishevelled, EGL-10 and Pleckstrin (DEP) Domain-Containing Protein DEPDC7 Binds to CARMA2 and CARMA3 Proteins, and Regulates NF-κB Activation
Egildo Luca D′ Andrea,Angela Ferravante,Ivan Scudiero,Tiziana Zotti,Carla Reale,Maddalena Pizzulo,Luigi R De La Motte,Chiara De Maio,Pellegrino Mazzone,Gianluca Telesio,Pasquale Vito,Romania Stilo +11 more
TL;DR: There is evidence that DEPDC7 may be required to specifically convey on the CBM complex signals coming from activated G protein-coupled receptors, and evidence that shRNA-mediated abrogation of DE PDC7 results in impaired NF-κB activation.
Journal ArticleDOI
CARMA2sh and ULK2 control pathogen-associated molecular patterns recognition in human keratinocytes: psoriasis-linked CARMA2sh mutants escape ULK2 censorship.
Ivan Scudiero,Pellegrino Mazzone,Luca E. D’Andrea,Angela Ferravante,Tiziana Zotti,Gianluca Telesio,Gabriele De Rubis,Carla Reale,Maddalena Pizzulo,Shanmugakonar Muralitharan,Pasquale Vito,Romania Stilo +11 more
TL;DR: It is demonstrated that in human keratinocytes CARMA2sh plays an essential role in the signal transduction pathway that connects pathogen-associated molecular patterns recognition to NF-κB activation, and a peptide blocking CARD-mediated BCL10 interactions reduces the capacity of psoriasis-linked CARma2sh mutants to activate NF-γB.
Journal ArticleDOI
Functional Characterization of Zebrafish (Danio rerio) Bcl10
Pellegrino Mazzone,Ivan Scudiero,Angela Ferravante,Marina Paolucci,Luca E. D’Andrea,Ettore Varricchio,Gianluca Telesio,Chiara De Maio,Maddalena Pizzulo,Tiziana Zotti,Carla Reale,Pasquale Vito,Romania Stilo +12 more
TL;DR: The functional characterization of a Danio rerio 241 amino acids polypeptide ortholog of the Caspase recruiting domain (CARD)-containing protein BCL10 shows that zebrafish may excellently serve as a model organism to study complex and intricate signal transduction pathways, such as those that control NF-κB activation.
Journal ArticleDOI
The E3 Ubiquitin Ligase RNF7 Negatively Regulates CARD14/CARMA2sh Signaling.
Gianluca Telesio,Ivan Scudiero,Maddalena Pizzulo,Pellegrino Mazzone,Tiziana Zotti,Serena Voccola,Immacolata Polvere,Pasquale Vito,Romania Stilo +8 more
TL;DR: Findings identify a new mechanism through which the ability of CARMA2 to activate NF-κB is regulated, which could have significant implications for the understanding of why mutations of this protein trigger human psoriasis.
Journal ArticleDOI
Antitumor activity of novel POLA1-HDAC11 dual inhibitors.
Sabrina Dallavalle,Loana Musso,Raffaella Cincinelli,Nadine Darwiche,Silvia Gervasoni,Giulio Vistoli,Mario B. Guglielmi,Ilaria La Porta,Maddalena Pizzulo,Elisa Modica,Federica Prosperi,Giacomo Signorino,Fabiana Colelli,Francesco Cardile,Alessandra Fucci,Egildo Luca D'Andrea,Assunta Riccio,Claudio Pisano +17 more
TL;DR: Hybrid molecules targeting simultaneously DNA polymerase α (POLA1) and histone deacetylases (HDACs) were designed and synthesized to exploit a potential synergy of action as discussed by the authors.
