Journal ArticleDOI
The akuB(KU80) mutant deficient for nonhomologous end joining is a powerful tool for analyzing pathogenicity in Aspergillus fumigatus.
Márcia Eliana da Silva Ferreira,Marcia Regina von Zeska Kress,Marcela Savoldi,Maria Helena S. Goldman,Albert Härtl,Thorsten Heinekamp,Axel A. Brakhage,Gustavo H. Goldman +7 more
TL;DR: To increase the frequency of homologous recombination, the KU80 homologue in Aspergillus fumigatus was inactivated and deletion had no influence on pathogenicity in a low-dose murine infection model.
Journal ArticleDOI
Expressed Sequence Tag Analysis of the Human Pathogen Paracoccidioides brasiliensis Yeast Phase: Identification of Putative Homologues of Candida albicans Virulence and Pathogenicity Genes
Gustavo H. Goldman,Everaldo dos Reis Marques,Diógenes Custódio Duarte Ribeiro,Luciano Ângelo de Souza Bernardes,Andréa Carla Quiapin,Patrícia Marostica Vitorelli,Marcela Savoldi,Camile P. Semighini,Regina Costa de Oliveira,Luiz R. Nunes,Luiz R. Travassos,Rosana Puccia,Wagner L. Batista,Leslie Ecker Ferreira,Júlio C. Moreira,Ana Paula Bogossian,Fredj Tekaia,Marina P. Nobrega,Francisco G. Nobrega,Maria Helena S. Goldman +19 more
TL;DR: The results suggest the possible conservation of pathogenicity and virulence mechanisms among fungi, expand considerably gene identification in P. brasiliensis, and provide a broader basis for further progress in understanding its biological peculiarities.
Journal ArticleDOI
Functional characterization of the Aspergillus fumigatus CRZ1 homologue, CrzA.
Frederico Marianetti Soriani,Iran Malavazi,Márcia Eliana da Silva Ferreira,Marcela Savoldi,Marcia Regina von Zeska Kress,Maria Helena S. Goldman,Omar Loss,Elaine Bignell,Gustavo H. Goldman +8 more
TL;DR: This work characterized an Aspergillus fumigatus CRZ1 homologue, CrzA and demonstrated its mediation of cellular tolerance to increased concentrations of calcium and manganese, and linked calcineurin activity with asexual developmental induction, finding that CrZA supports appropriate developmental induction in a calcineURin and brlA‐dependent manner in both species.
Journal ArticleDOI
Transcriptome analysis of Aspergillus fumigatus exposed to voriconazole
Márcia Eliana da Silva Ferreira,Iran Malavazi,Marcela Savoldi,Axel A. Brakhage,Maria Helena S. Goldman,H. Stanley Kim,William C. Nierman,Gustavo H. Goldman +7 more
TL;DR: A decreased mRNA expression of various ergosterol biosynthesis genes; increased mRNA levels of genes involved in a variety of cell functions, such as transporters, transcription factors, proteins involved in cell metabolism, and hypothetical proteins; and the involvement of the cyclic AMP-protein kinase signaling pathway in the increased expression of several of these genes.
Journal ArticleDOI
Functional characterization of an Aspergillus fumigatus calcium transporter (PmcA) that is essential for fungal infection.
Taisa Magnani Dinamarco,Fernanda Zanolli Freitas,Ricardo Almeida,Neil Andrew Brown,Thaila Fernanda dos Reis,Leandra Naira Zambelli Ramalho,Marcela Savoldi,Maria Helena S. Goldman,Maria Célia Bertolini,Gustavo H. Goldman,Gustavo H. Goldman +10 more
TL;DR: It is demonstrated that CrzA can directly control the mRNA accumulation of the pmcA-C genes by binding to their promoter regions, supporting the interaction between calcineurin and the function of these transporters.
