Journal ArticleDOI
NLRP3 inflammasomes are required for atherogenesis and activated by cholesterol crystals
Peter Duewell,Hajime Kono,Katey J. Rayner,Katey J. Rayner,Cherilyn M. Sirois,Gregory I. Vladimer,Franz Bauernfeind,George S. Abela,Luigi Franchi,Guillermo Gabriel Nuñez,Max Schnurr,Terje Espevik,Egil Lien,Katherine A. Fitzgerald,Kenneth L. Rock,Kathryn J. Moore,Kathryn J. Moore,Samuel D. Wright,Veit Hornung,Eicke Latz,Eicke Latz +20 more
TL;DR: It is shown that cholesterol crystals activate the NLRP3 inflammasome in phagocytes in vitro in a process that involves phagolysosomal damage and that crystalline cholesterol acts as an endogenous danger signal and its deposition in arteries or elsewhere is an early cause rather than a late consequence of inflammation.
Journal ArticleDOI
Colitis induced in mice with dextran sulfate sodium (DSS) is mediated by the NLRP3 inflammasome
Christian Bauer,Peter Duewell,Christine Mayer,Hans A. Lehr,Katherine A. Fitzgerald,Marc Dauer,Jürg Tschopp,Stefan Endres,Eicke Latz,Max Schnurr +9 more
TL;DR: The NLRP3 inflammasome was identified as a critical mechanism of intestinal inflammation in the DSS colitis model and may serve as a potential target for the development of novel therapeutics for patients with IBD.
Journal ArticleDOI
Therapeutic Efficacy of Bifunctional siRNA Combining TGF-β1 Silencing with RIG-I Activation in Pancreatic Cancer
Jonathan Ellermeier,Jiwu Wei,Peter Duewell,Sabine Hoves,Mareike R Stieg,Tina Adunka,Daniel Noerenberg,Hans-Joachim Anders,Doris Mayr,Hendrik Poeck,Gunther Hartmann,Stefan Endres,Max Schnurr +12 more
TL;DR: In this article, a bifunctional ppp-siRNA that combines RIG-I activation with gene silencing of TGF-β1 (ppp-TGF-α) was presented.
Journal ArticleDOI
Colitis induced in mice with dextran sulfate sodium (DSS) is mediated by the NLRP3 inflammasome
Journal ArticleDOI
RIG-I-like helicases induce immunogenic cell death of pancreatic cancer cells and sensitize tumors toward killing by CD8(+) T cells.
Peter Duewell,A Steger,H Lohr,H Bourhis,H Hoelz,S V Kirchleitner,M R Stieg,S Grassmann,Sebastian Kobold,Jens T. Siveke,Stefan Endres,Max Schnurr,Max Schnurr +12 more
TL;DR: RLH ligands induce a highly immunogenic form of tumor cell death linking innate and adaptive immunity, and MDA5-based immunotherapy led to effective tumor control of established pancreatic tumors.
