The present study shows that recombinant interleukin 2 (IL-2) purified to homogeneity induces a rapid and potent enhancement of spontaneous cytotoxicity of human peripheral blood lymphocytes. The cells mediating cytotoxicity after 18-h treatment with IL-2 have surface markers of natural killer (NK) cells and are generated from the peripheral blood subset containing spontaneous cytotoxic cells. A parallel production of gamma interferon (IFN-gamma) is induced by recombinant IL-2 (rIL-2), and NK cells appear to be the major producer cells, whereas T cells are unable to produce IFN-gamma under these experimental conditions. However, the kinetics of the enhancement of cytotoxicity are faster than those of IFN-gamma production, and monoclonal anti-IFN-gamma antibodies do not suppress this effect, making it unlikely that the IFN-gamma produced is responsible for the enhancement. The enhancement of NK cell activity induced by rIL-2 precedes any proliferative response of the lymphocytes, which is instead observed in longer-term cultures of both NK and T cells.

Response of resting human peripheral blood natural killer cells to interleukin 2.

Recent research on empathy in humans and other mammals seeks to dissociate emotional and cognitive empathy. These forms, however, remain interconnected in evolution, across species and at the level of neural mechanisms. New data have facilitated the development of empathy models such as the perception-action model (PAM) and mirror-neuron theories. According to the PAM, the emotional states of others are understood through personal, embodied representations that allow empathy and accuracy to increase based on the observer's past experiences. In this Review, we discuss the latest evidence from studies carried out across a wide range of species, including studies on yawn contagion, consolation, aid-giving and contagious physiological affect, and we summarize neuroscientific data on representations related to another's state.

Mammalian empathy: behavioural manifestations and neural basis

Interleukin 2 (IL 2) (Aarden et at. 1979) is a genetically unrestricted, soluble immunoenhancing factor which is produced by helper T cells following stimulation with either T cell mitogens or allogeneic cells. Presumably, IL 2 is also produced and secreted following specific antigen stimulation, although this question has not been carefully evaluated. The definitive biological assay for IL 2 measures the factor's ability to maintain the growth of factor-dependent cytotoxic T cell lines. However, a broad spectrum of biological activities has been ascribed to IL 2. The factor has been reported to induce thymocyte proliferation either in the presence (Chen & DiSabato 1976, Paetkau et al. 1976) or absence (Farrar et al. 1978) of a suboptimal concentration of T cell mitogen. Additionally, IL 2 has been shown to augment the proliferation and generation of cytotoxic cells by alloantigenstimulated T cell populations (Wagner & Rollinghoff 1978, Farrar et al. 1978) and, in the process, induce the synthesis of immune interferon (IFNji) by the alloantigen-stimulated T cells (Farrar et al. 1981a). The murine immune interferon (IFN-gamma) gene was cloned and expressed under control of the simian virus 40 early promoter in the monkey COS-1 cell line. A protein is secreted from these cells having the biological, antigenic, and biochemical characteristics of natural murine IFN-gamma. Cloned murine IFN-gamma cDNAs were obtained by using RNA from both mitogen-induced murine spleens and the transfected COS cells, and both code for identical proteins. The mature murine IFN-gamma encoded is 136 amino acids long, 10 amino acids shorter than human IFN-gamma. The nucleotide homology between the murine and human IFN-gamma genes is 60-65%, whereas the encoded proteins are only 40% homologous. Murine IFN-gamma cDNA was expressed in Escherichia coli under trp promoter control.

Cloning and expression of murine immune interferon cDNA.

By three criteria, we have demonstrated that B cell stimulatory factor (BSF-1) and B cell differentiation factor (BCDF-gamma) are the same lymphokine. Highly purified preparations of high performance liquid chromatography-purified or affinity-purified BSF-1 had BCDF-gamma activity but not BCDF-mu activity. A monoclonal anti-BSF-1 antibody coupled to Sepharose depleted both BSF-1 and BCDF-gamma activity but not BCDF-mu activity from two different T cell supernatants. Soluble monoclonal anti-BSF-1 blocked the BSF-1 and BCDF-gamma but not the BCDF-mu responses. These results suggest that BSF-1 acts on both resting and activated B cells to induce different effects.

SEROLOGICAL, BIOCHEMICAL, AND FUNCTIONAL IDENTITY OF B CELL-STIMULATORY FACTOR 1 AND B CELL DIFFERENTIATION FACTOR FOR IgG1

Observations of chimpanzees (Pan troglodytes) and bonobos (Pan paniscus) provide valuable comparative data for understanding the significance of conspecific killing. Two kinds of hypothesis have been proposed. Lethal violence is sometimes concluded to be the result of adaptive strategies, such that killers ultimately gain fitness benefits by increasing their access to resources such as food or mates. Alternatively, it could be a non-adaptive result of human impacts, such as habitat change or food provisioning. To discriminate between these hypotheses we compiled information from 18 chimpanzee communities and 4 bonobo communities studied over five decades. Our data include 152 killings (n = 58 observed, 41 inferred, and 53 suspected killings) by chimpanzees in 15 communities and one suspected killing by bonobos. We found that males were the most frequent attackers (92% of participants) and victims (73%); most killings (66%) involved intercommunity attacks; and attackers greatly outnumbered their victims (median 8:1 ratio). Variation in killing rates was unrelated to measures of human impacts. Our results are compatible with previously proposed adaptive explanations for killing by chimpanzees, whereas the human impact hypothesis is not supported.

Production of leukocyte interferon (IFN-alpha) and fibroblast interferon (IFN-beta) can be induced by a variety of agents but immune interferon, IFN-gamma, is only obtained when lymphoid cells are stimulated by specific antigens, allo-antigens or T-cell mitogens. Moreover, in bulk cultures, only small quantities of IFN-gamma are produced. The type of cell producing IFN-gamma has not been unambiguously defined and so we set out to determine whether a pure T-cell population could produce it, exploiting the knowledge that T cells can be maintained indefinitely in tissue culture by the addition of T-cell growth factors. Although not all T cells can found long-term cultures of this kind, cultures of antigen-specific helper, suppressor and killer T cells have been obtained in this way. We now describe the production of substantial amounts of INF-gamma when some (but not all) murine T-cell clones derived from such cultures are stimulated by either concanavalin A (Con A) or phytohaemagglutinin (PHA).

Production of immune interferon by murine T-cell clones from long-term cultures.

Chimpanzees (Pan troglodytes) make nests for resting and sleeping, which is unusual for anthropoid primates but common to all great apes. Arboreal nesting has been linked to predation pressure, but few studies have tested the adaptive nature of this behavior. We collected data at two chimpanzee study sites in southeastern Senegal that differed in predator presence to test the hypothesis that elevated sleeping platforms are adaptations for predator defense. At Assirik in the Parc National du Niokolo-Koba, chimpanzees face four species of large carnivore, whereas at Fongoli, outside national park boundaries, humans have exterminated almost all natural predators. We quantified the availability of vegetation at the two sites to test the alternative hypothesis that differences in nesting reflect differences in habitat structure. We also examined possible sex differences in nesting behavior, community demographic differences, seasonality and nest age differences as variables also potentially affecting nest characteristics and nesting behavior between the two sites. Chimpanzees at Fongoli nested at lower heights and farther apart than did chimpanzees at Assirik and sometimes made nests on the ground. The absence of predators outside of the national park may account for the differences in nest characteristics at the two sites, given the similarities in habitat structure between Fongoli and Assirik. However, Fongoli chimpanzees regularly build arboreal nests for sleeping, even under minimal predation pressure, and this requires explanation. Am. J.
