Journal ArticleDOI

Establishment of sink conditions in dissolution rate determinations. Theoretical considerations and application to nondisintegrating dosage forms

TL;DR: It is shown that unless sink conditions are maintained, in vitro results will bear little relationship to in vivo observations and the use of an organic solvent phase which can function as a reservoir for dissolved drug and maintain sink conditions in the aqueous dissolution fluid is demonstrated.
Journal ArticleDOI

Influence of first‐pass effect on availability of drugs on oral administration

TL;DR: Plasma level data for propranolol in man have been used to indicate the utility of these equations, and the significance of these calculations to the design of clinical studies with new drugs intended for oral use is discussed.
Journal ArticleDOI

Increasing dissolution rates and gastrointestinal absorption of drugs via solid solutions and eutectic mixtures I. Theoretical considerations and discussion of the literature

TL;DR: This report proposes that results previously attributed to eutectic mixtures are properly explained by the existence of solid solutions and a mechanism is considered by which such solid systems may enhance dissolution rates and, in turn, the gastrointestinal absorption rate and availability of poorly soluble drugs.
