Journal ArticleDOI
A conserved dendritic-cell regulatory program limits antitumour immunity
Barbara Maier,Andrew Leader,Steven T. Chen,Navpreet Tung,Christie Chang,Jessica LeBerichel,Aleksey Chudnovskiy,Shrisha Maskey,Laura Walker,John P. Finnigan,Margaret E. Kirkling,Margaret E. Kirkling,Boris Reizis,Sourav Ghosh,Natalie Roy D’Amore,Nina Bhardwaj,Carla V. Rothlin,Andrea S. Wolf,Raja M. Flores,Thomas U. Marron,Adeeb Rahman,Ephraim Kenigsberg,Brian D. Brown,Miriam Merad +23 more
TL;DR: A regulatory module associated with tumour-antigen uptake that reduces DC1 functionality in human and mouse cancers is uncovered and modulating this program can rescue antitumor immunity in mice.
Journal ArticleDOI
Single-cell analysis of human non-small cell lung cancer lesions refines tumor classification and patient stratification.
Andrew Leader,John A. Grout,Barbara Maier,Barzin Y. Nabet,Matthew D. Park,Alexandra Tabachnikova,Christie Chang,Laura Walker,Alona Lansky,Jessica Le Berichel,Leanna Troncoso,Nausicaa Malissen,Melanie Davila,Jerome Martin,Giuliana Magri,Kevin Tuballes,Zhen Zhao,Francesca Petralia,Robert M. Samstein,Natalie Roy D’Amore,Gavin Thurston,Alice O. Kamphorst,Andrea S. Wolf,Raja M. Flores,Pei Wang,Sören Müller,Ira Mellman,Mary Beth Beasley,Hélène Salmon,Adeeb Rahman,Thomas U. Marron,Ephraim Kenigsberg,Miriam Merad +32 more
TL;DR: In this article, a cellular module consisting of PDCD1+CXCL13+ activated T cells, IgG+ plasma cells, and SPP1+ macrophages, referred to as the lung cancer activation module (LCAMhi), was identified.
Journal ArticleDOI
Identification of a lung cancer cell line deficient in atg7-dependent autophagy
Jonathan Mandelbaum,Neil Rollins,Pooja Shah,Doug Bowman,Janice Y Lee,Olga Tayber,Hugues Bernard,Patrick J. LeRoy,Ping Li,Erik Koenig,James E. Brownell,Natalie Roy D’Amore +11 more
TL;DR: This study identified a rare case of a cancer cell line, H1650 lung adenocarcinoma, which has lost ATG7 expression due to a focal biallelic deletion within the ATG 7 locus, and characterized several phenotypes reported to be influenced by autophagy, and observed an AtG7-dependent increase in cell growth and clearance of proteasome-inhibitor induced protein aggregates.
Journal ArticleDOI
Plk1 inhibition causes post-mitotic DNA damage and senescence in a range of human tumor cell lines.
Denise L. Driscoll,Arijit Chakravarty,Doug Bowman,Vaishali Shinde,Kerri Lasky,Judy Shi,Tricia J. Vos,Bradley Stringer,Ben Amidon,Natalie Roy D’Amore,Marc L. Hyer +10 more
TL;DR: It is demonstrated for the first time that cellular senescence is the predominant outcome of Plk1 inhibition in some cancer cell Lines, whereas in other cancer cell lines the dominant outcome appears to be apoptosis, as has been reported in the literature.
Posted ContentDOI
CITEseq analysis of non-small-cell lung cancer lesions reveals an axis of immune cell activation associated with tumor antigen load and TP53 mutations
Andrew Leader,John A. Grout,Christie Chang,Barbara Maier,Alexandra Tabachnikova,Laura Walker,Alona Lansky,Jessica LeBerichel,Naussica Malissen,Melanie Davila,Jerome Martin,Giuliana Magri,Kevin Tuballes,Zhen Zhao,Francesca Petralia,Robert M. Samstein,Natalie Roy D’Amore,Gavin Thurston,Alice O. Kamphorst,Andrea S. Wolf,Raja M. Flores,Pei Wang,Mary Beth Beasley,Hélène Salmon,Adeeb Rahman,Thomas U. Marron,Ephraim Kenigsberg,Miriam Merad +27 more
TL;DR: The data establish LCAM as a key mode of immune cell activation associated with high tumor antigen load and driver mutations in smokers and non-smokers.
