Journal Article

Taxol metabolism by human liver microsomes: identification of cytochrome P450 isozymes involved in its biotransformation

TL;DR: Microsomes from patients treated with barbiturates and benzodiazepines increased the formation of metabolite M4 to the level of metabolites M5, demonstrating that drug interactions could modify the human metabolism of taxol.
Journal ArticleDOI

Regioselective Metabolism of Taxoids by Human CYP3A4 and 2C8: Structure-Activity Relationship

TL;DR: Data strongly suggested that the structure of the lateral chain and the nature of substituent in position 10 play an important role in determining the regioselective oxidation by P450 proteins and modulate the reaction rate by human liver microsomes.
Journal Article

Hepatic metabolism and biliary excretion of Taxol in rats and humans.

TL;DR: The biliary excretion of Taxol has been studied in rats and in a human patient receiving Taxol in a phase I trial, and the chemical structures of four of the rat metabolites and three of the human metabolites have been identified.
Journal Article

Modification of Paclitaxel Metabolism in a Cancer Patient by Induction of Cytochrome P450 3A4

TL;DR: A high CYP3A4 activity in the patient is consistent with the repeated administration of methylprednisolone for 14 days before paclitaxel treatment, a compound known to induce the CYP1A isoform, and with the increased ratio of 6beta-hydroxycortisol/ cortisol in urine, an index of CYP2A activity.
