Journal ArticleDOI
New mass spectrometric methods for the study of noncovalent associations of biopolymers
TL;DR: The potential for high throughput affinity screening of combinatorial libraries using ESI–MS is described based upon the multi-stage MS capability of Fourier transform ion cyclotron resonance instrumentation and involving the characterization of components of the library constituents initially present as complexes with a target biopolymer in the ion trap.
Journal ArticleDOI
Probing the energetics of dissociation of carbonic anhydrase-ligand complexes in the gas phase.
Jinming Gao,Qinyuan Wu,Jeffrey D. Carbeck,Q. Paula Lei,Richard D. Smith,George M. Whitesides +5 more
TL;DR: Results indicate that specific interactions between the sulfonamide group on the inhibitor and the Zn(II) ion on CAII were preserved in the gas phase, consistent with the hypothesis that CAII retains, at least partially, the structure of its binding pocket in theGas phase on the time scale (seconds to minutes) of the ESI-FTICR measurements.
Journal ArticleDOI
Tyrosine O-sulfation proteoforms affect HIV-1 monoclonal antibody potency
Cindy X. Cai,Nicole A. Doria-Rose,Nicole A. Schneck,Vera B. Ivleva,Bradford W Tippett,William R. Shadrick,Sarah O’Connell,Jonathan W. Cooper,Zachary Schneiderman,Bao Shan Zhang,Daniel B. Gowetski,Daniel Blackstock,Jacob Demirji,Bob C. Lin,Jason Gorman,Tracy L. McGaha Liu,Yile Li,Adrian B. McDermott,Peter D. Kwong,Kevin Carlton,Jason G. D. Gall,Q. Paula Lei +21 more
TL;DR: In this paper , the authors developed a hydrophobic interaction chromatography (HIC) method to separate and quantify different sulfation proteoforms, which led to the direct functionality assessment of tyrosine-sulfated species.
Journal ArticleDOI
Advances in purification of SARS-CoV-2 spike ectodomain protein using high-throughput screening and non-affinity methods
Nicole L. Cibelli,G. Arias,McKenzie L. Figur,Shireen Khayat,Kristin M. Leach,Ivan Loukinov,William R. Shadrick,Watchalee Chuenchor,Yaroslav Tsybovsky,Aakash Patel,Misa Mai,Xin Wang,Karen Vickery,Tina Khin,Renata Skubutyte,Farah Vejzagic,Niutish Bastani,A.A. Vinitsky,Q. Paula Lei,Krishana C. Gulla,Daniel B. Gowetski +20 more
TL;DR: In this paper , the first known non-affinity purification method for two S constructs, S_dF_2P and HexaPro, expressed in the mammalian cell line, CHO-DG44, was presented.
Journal ArticleDOI
Analytical characterization of broadly neutralizing antibody CAP256LS heavy chain clipping during manufacturing development
Deepika Gollapudi,Erwin Rosales-Zavala,Vera B. Ivleva,Yanhong Yang,Yile Li,Sarah E O'Connell,Nicole A. Doria-Rose,Aakash Patel,Daniel Blackstock,Daniel B. Gowetski,Kevin Carlton,Jason G. D. Gall,Q. Paula Lei +12 more
TL;DR: How analytical characterization tools, especially the microchip reduced capillary gel electrophoresis (CGE-SDS), played a pivotal role in identifying the development issues and in providing measurements to guide implementation of mitigation strategies is detailed.
