Power analysis of transcriptome-wide association study: Implications for practical protocol choice

TL;DR: In this paper, the authors compared the power of GWAS, TWAS, and a hypothetical protocol utilizing real expression data and found that the use of predicted expressions in TWAS is superior to actual expressions.
Posted ContentDOI

Power analysis of transcriptome-wide association study

TL;DR: It is found that depending on the type of relationship between genetics, gene expression, and disease, the estimates used by TWAS could be actually more relevant than real gene expressions, and that TWAS is not always better than GWAS when the relationship between Genetics and expression is weak.

Power analysis of transcriptome-wide association study: implications for practical protocol choice

TL;DR: In this article, the power of transcriptome-wide association study (TWAS) was compared with standard Genome-Wide Association Study (GWAS) and expression medicated GWAS (emGWAS), with respect to two representative scenarios: causality and pleiotropy.

Disentangling genetic feature selection and aggregation in transcriptome-wide association studies

TL;DR: It is demonstrated that current linear models of GReX inadvertently combine two separable steps of machine learning - feature selection and aggregation - which can be independently replaced to improve overall power.
