Structure and Dynamics of a 197 bp Nucleosome in Complex with Linker Histone H1
Jan Bednar,Isabel Garcia-Saez,Ramachandran Boopathi,Amber R. Cutter,Gabor Papai,Anna Reymer,Sajad Hussain Syed,Sajad Hussain Syed,Imtiaz Nisar Lone,Imtiaz Nisar Lone,Ognyan Tonchev,Ognyan Tonchev,Corinne Crucifix,Hervé Menoni,Hervé Menoni,Christophe Papin,Dimitrios A. Skoufias,Hitoshi Kurumizaka,Richard Lavery,Ali Hamiche,Jeffrey J. Hayes,Patrick Schultz,Dimitar Angelov,Carlo Petosa,Stefan Dimitrov +24 more
TL;DR: Findings reveal that H1 imparts a strong degree of asymmetry to the nucleosome, which is likely to influence the assembly and architecture of higher-order structures.
Journal ArticleDOI
Structure of an H1-Bound 6-Nucleosome Array Reveals an Untwisted Two-Start Chromatin Fiber Conformation
Isabel Garcia-Saez,Hervé Menoni,Hervé Menoni,Ramachandran Boopathi,Ramachandran Boopathi,Manu S. Shukla,Manu S. Shukla,Lama Soueidan,Lama Soueidan,Marjolaine Noirclerc-Savoye,Aline Le Roy,Dimitrios A. Skoufias,Jan Bednar,Jan Bednar,Ali Hamiche,Dimitar Angelov,Carlo Petosa,Stefan Dimitrov,Stefan Dimitrov +18 more
TL;DR: In this article, the crystal structure of a 6-nucleosome array bound to linker histone H1 was determined under ionic conditions that favor incomplete chromatin condensation.
Journal ArticleDOI
The Flexible Ends of CENP-A Nucleosome Are Required for Mitotic Fidelity
Yohan Roulland,Khalid Ouararhni,Mladen Naidenov,Lorrie Ramos,Muhammad Shuaib,Sajad Hussain Syed,Sajad Hussain Syed,Imtiaz Nizar Lone,Imtiaz Nizar Lone,Ramachandran Boopathi,Ramachandran Boopathi,Emeline Fontaine,Gabor Papai,Hiroaki Tachiwana,Thierry Gautier,Dimitrios A. Skoufias,Kiran Padmanabhan,Jan Bednar,Hitoshi Kurumizaka,Patrick Schultz,Dimitar Angelov,Ali Hamiche,Ali Hamiche,Stefan Dimitrov +23 more
TL;DR: The data reveal that the evolutionarily conserved flexible ends of the CENP-A nucleosomes are essential to ensure the fidelity of the mitotic pathway.
Journal ArticleDOI

Skoufias,Hitoshi Kurumizaka,Richard Lavery,Ali Hamiche,Jeffrey J. Hayes,Patrick Schultz,Dimitar Angelov,Carlo Petosa,Stefan Dimitrov +20 more
Journal ArticleDOI
Phase-plate cryo-EM structure of the Widom 601 CENP-A nucleosome core particle reveals differential flexibility of the DNA ends.
Ramachandran Boopathi,Radostin Danev,Maryam Khoshouei,Seyit Kale,Seyit Kale,Sunil Nahata,Lorrie Ramos,Dimitar Angelov,Stefan Dimitrov,Stefan Dimitrov,Ali Hamiche,Carlo Petosa,Jan Bednar,Jan Bednar +13 more
TL;DR: The cryo-EM structure of the CENP-A 601 NCP determined by Volta phase-plate imaging reveals that one (‘left’) 601 DNA end is well ordered whereas the other end is flexible and partly detached from the histone core, suggesting sequence-dependent dynamics of the DNA termini.
