Selenium (Se) is an essential micronutrient for humans and animals, but lead to toxicity when taken in excessive amounts. Plants are the main source of dietary Se, but essentiality of Se for plants is still controversial. However, Se at low doses protects the plants from variety of abiotic stresses such as cold, drought, desiccation and metal stress. In animals, Se acts as an antioxidant and helps in reproduction, immune responses, thyroid hormone metabolism. Selenium is chemically similar to sulfur, hence taken up inside the plants via sulfur transporters present inside root plasma membrane, metabolized via sulfur assimilatory pathway, and volatilized into atmosphere. Selenium induced oxidative stress, distorted protein structure and function, are the main causes of Se toxicity in plants at high doses. Plants can play vital role in overcoming Se deficiency and Se toxicity in different regions of the world, hence, detailed mechanism of Se metabolism inside the plants is necessary for designing effective Se phytoremediation and biofortification strategies.

An Overview of Selenium Uptake, Metabolism, and Toxicity in Plants.

Selenium is regulated in the body to maintain vital selenoproteins and to avoid toxicity. When selenium is limiting, cells utilize it to synthesize the selenoproteins most important to them, creating a selenoprotein hierarchy in the cell. The liver is the central organ for selenium regulation and produces excretory selenium forms to regulate whole-body selenium. It responds to selenium deficiency by curtailing excretion and secreting selenoprotein P (Sepp1) into the plasma at the expense of its intracellular selenoproteins. Plasma Sepp1 is distributed to tissues in relation to their expression of the Sepp1 receptor apolipoprotein E receptor-2, creating a tissue selenium hierarchy. N-terminal Sepp1 forms are taken up in the renal proximal tubule by another receptor, megalin. Thus, the regulated whole-body pool of selenium is shifted to needy cells and then to vital selenoproteins in them to supply selenium where it is needed, creating a whole-body selenoprotein hierarchy.

Regulation of Selenium Metabolism and Transport

In recent years, ultrasound-assisted extraction (UAE) has attracted growing interest, as it is an effective method for the rapid extraction of a number of compounds from food and environmental samples, with extraction efficiency comparable to that of classical techniques. In particular, recently, numerous analytical applications of this technique dealt with the extraction of natural compounds and pollutants from food and environmental samples. This review gives a brief presentation of the theory of UAE, discusses recent advances that influence its efficiency, and summarizes the main results in selected applications published in the period 2010–12. There is discussion of the advantages and the disadvantages of UAE and the possibility of coupling UAE with other analytical techniques.

Selenium is an essential immunonutrient which holds the human's metabolic activity with its chemical bonds. The organic forms of selenium naturally present in human body are selenocysteine and selenoproteins. These forms have a unique way of synthesis and translational coding. Selenoproteins act as antioxidant warriors for thyroid regulation, male-fertility enhancement, and anti-inflammatory actions. They also participate indirectly in the mechanism of wound healing as oxidative stress reducers. Glutathione peroxidase (GPX) is the major selenoprotein present in the human body, which assists in the control of excessive production of free radical at the site of inflammation. Other than GPX, other selenoproteins include selenoprotein-S that regulates the inflammatory cytokines and selenoprotein-P that serves as an inducer of homeostasis. Previously, reports were mainly focused on the cellular and molecular mechanism of wound healing with reference to various animal models and cell lines. In this review, the role of selenium and its possible routes in translational decoding of selenocysteine, synthesis of selenoproteins, systemic action of selenoproteins and their indirect assimilation in the process of wound healing are explained in detail. Some of the selenium containing compounds which can acts as cancer preventive and therapeutics are also discussed. These compounds directly or indirectly exhibit antioxidant properties which can sustain the intracellular redox status and these activities protect the healthy cells from reactive oxygen species induced oxidative damage. Although the review covers the importance of selenium/selenoproteins in wound healing process, still some unresolved mystery persists which may be resolved in near future.

Selenium and selenoproteins: it's role in regulation of inflammation.

Wheat (Triticum aestivum) collected in the Nawanshahr-Hoshiarpur Region (Punjab, India) showed the highest selenium concentrations ever recorded in cereal grains (29−185 μg g−1). There was a strong positive relationship between the selenium content in shoots and that in kernels, showing that grain selenium concentration can be predicted from that in the vegetative tissues of the plant. The identity and content of the selenocompounds in the grain samples and in wheat-based reference materials were investigated by HPLC−ICP−dynamic reaction cell−MS. Reversed-phase, cation exchange, and anion exchange HPLC were used to separate the selenium species after ultrasound-assisted enzymatic extraction with an ultrasonic probe. Selenomethionine and selenate accounted for 72−85% and 2−6% of the sum of the selenium species, respectively.
