Genetic profiling: Differentiation and identification of hazelnut cultivars (Corylus avellana L.) using RAPD-PCR

The effects of cultural conditions on the mycelial growth and fruit-body formation and the yield of Pleurotus sajor-caju grown in rice straw medium were examined. The change of content of chemical components of mushroom and the medium during the cultivation period was also examined. The growth of mycelium was best in the medium sterilized for 60 min at with a moisture content of 60%. The optimum pH and temperature for mycelial growth were 6.0 and , respectively, whereas the optimum pH and temperature for fruit-body formation ranged from 5.0 to 6.0 and from 20 to , respectively. Rice bran and starch cake were better than the other additives used for both mycelial growth and fruit-body formation. The optimum concentration of rice bran in the medium was 10%. The total yield of mushroom was , 66% of which was prouduced from the first and second cropping period. As the harvest proceeded, the crude fiber cotent increased in the mushroom, whereas it decreased in the medium. The content of free sugar and crude ash, on the other hand, decreased in the mushroom, whereas it increased in the medium. The content of the other chemical components examined decreased in both the mush­room and the medium.

Studies on Cultivation of Pleurotus sajor-caju (I) -Cultural Conditions and Changes of Chemical Components-

This investigation was conducted at the Experimental Farm, Fac. Tech. and Develop. (Gazala region – Zagazig), Zagazig Univ., Sharkia Governorate under open field during the two fall – winter seasons of 2015/2016 and 2016/2017, to study the effect of three cultivars of squash ( Eskandarany, Revera and Azyad) and foliar spray treatments with, yeast extract at the rate of 3 and 6gl-1 ,as well as potassium silicate at the rate of 2 and 4gl-1 plus control (tap water)and their interactions on flowering expression, i.e., number of male and female flowers plant-1, sex ratio and femalness percentage, yield and its components (number of fruits  plant-1 , fruit fresh weight, and dry matter percentage, total fruit yield plot-1 and relative yield), as well as fruit quality, i.e., fruit physical characters (both of fruit length and diameter) and fruit chemical composition (nitrogen, phosphorus and potassium content, protein and total carbohydrates percentage). Results showed the superiority of the two tested cultivars Azyad and Revera at all studied characteristics compared with Eskandarany cv. Spraying  with yeast extract or potassium silicate significantly increased most parameters of flowering, fruit yield and quality attributes of squash than control treatment . Moreover, spraying plants with potassium silicate at rate of 4gl-1 or yeast extract  at 6gl-1 significantly surpassed compared with the other foliar treatments and control in this respect. The best results of the interaction treatments for flowering, yield and fruit parameters were recorded by the two cultivars Azyad or Revera combined with the foliar spray by 4gl-1 of potassium silicate or 6gl-1 of yeast extract which recorded increased in most of all the mentioned studied parameters. Hence, each of these treatments could be recommended for enhancement flowering, yield and fruit quality of squash plants with low cost production when planted in open field at fall – winter season under the same conditions of this experiment.

Influence of some squash cultivars and growth stimulants on flowering, yield and fruit quality at autumn – winter season under open field conditions

Clusterbean (Cyamopsis tetragonoloba (L.) Taub.), also known as Guar is a widely cultivated dryland legume of Western India and parts of Africa. Apart from being a vegetable crop, it is also an abundant source of a natural hetero-polysaccharide called guar gum or galactomannan which is widely used in cosmetics, pharmaceuticals, food processing, shale gas drilling, etc. Here, for the first time we are reporting a chromosome-scale reference genome assembly of clusterbean, from a high galactomannan containing popular guar cultivar, RGC-936, by combining sequenced reads from Illumina, 10x Chromium and Oxford Nanopore technologies. The initial assembly of 1580 scaffolds with an N50 value of 7.12 Mbp was generated. Then, the final genome assembly was obtained by anchoring these scaffolds to a high density SNP map. Finally, a genome assembly of 550.31 Mbp was obtained in 7 pseudomolecules corresponding to 7 chromosomes with a very high N50 of 78.27 Mbp. We finally predicted 34,680 protein-coding genes in the guar genome. The high-quality chromosome-scale cluster bean genome assembly will facilitate understanding of the molecular basis of galactomannan biosynthesis and aid in genomics-assisted breeding of superior cultivars.

Summer Squash (Cucurbita pepo L.) locally known as „chappankaddoo‟ or „valyatikaddoo‟ belongs to family cucurbitaceae is a native of Mexico and US. In India, Uttar Pradesh, Punjab and Haryana are the states where summer squash is considered among commonly consumed vegetable crops (Dhaliwal, 2012) with cultivation limits extending to Himachal Pradesh, Jammu and Kashmir and West Bengal. It is tender, annual, warm season vegetable crop that is harvested when fruits are immature (Sarhan et al., 2011) and contains carbohydrates, fibre, potassium, vitamin B and vitamin C that makes it higher in food value. India share 14.78% production of squashes, pumpkins and gourds to worlds total production and stands at second position after China (22.83%) (Anonymous, 2016).

Effect of Mulching on Growth, Yield and Quality in Different Varieties of Summer Squash (Cucurbita pepo L.)

High temperature affects diverse physiological, biochemical and molecular processes including gene expression and genetic stability. To demostrate this, primary leaves of moth bean were harvested from eight days old seedlings followed by heat shock treatment at 37, 42, 47 and 52 °C for 10, 20 and 30 min. The response of tissue for callusing was comparatively early in explants treated at 37, 42 and 47 °C for 10 min. However, regeneration was negatively affected by most of the heat treatments. A few polypeptides were found to be up regulated as well as down regulated with heat shock treatments. Some proteins were specifically regulated at higher temperatures of 42° and 47 °C. Two polypeptides were also up regulated in the protein profiling of callus; however, these were different than the once observed in protein profiling of leaf explant immediately after heat treatment. Moreover, these bands were found in only one treatment each, viz. one in 20 min of 47 °C (100 kD) and another in 30 min of 47 °C (36.7 kD). These may be expected to be consequence of genetic change (mutation). RAPD analysis further revealed that plantlets obtained at 47 °C generated a novel band indicating mutagenic effect of heat shock treatment.

Pleurotus is generally known as Oyster mushroom all over the world and Dhingri in India (Lovkesh et al., 2006). Mushroom has been defined as a macro-fungus with a distinctive fruiting body, which can be epigeous or hypogenous, large enough to be seen with the naked eye and to be picked by hand (Chang and Miles, 1989). Oyster mushroom is one of the most popular edible mushroom and belong to the genus Pleurotus and the family Pleurotaceae. Pleurotus was first cultivated in Germany as a subsistence measure during World War I (Flack, 1917) and is now grown commercially around the world for food. Oyster mushroom is one of the most commonly sought wild mushrooms, though it can also be cultivated on straw and other media. Pleurotus mushrooms are primary decomposers of hardwood trees and are found worldwide.
