The Role of Oxidative Stress in the Pathogenesis of Age-Related Macular Degeneration

The development of cancer in humans and animals is a multistep process. The complex series of cellular and molecular changes participating in cancer development are mediated by a diversity of endogenous and exogenous stimuli. One type of endogenous damage is that arising from intermediates of oxygen (dioxygen) reduction - oxygen-free radicals (OFR), which attacks not only the bases but also the deoxyribosyl backbone of DNA. Thanks to improvements in analytical techniques, a major achievement in the understanding of carcinogenesis in the past two decades has been the identification and quantification of various adducts of OFR with DNA. OFR are also known to attack other cellular components such as lipids, leaving behind reactive species that in turn can couple to DNA bases. Endogenous DNA lesions are genotoxic and induce mutations. The most extensively studied lesion is the formation of 8-OH-dG. This lesion is important because it is relatively easily formed and is mutagenic and therefore is a potential biomarker of carcinogenesis. Mutations that may arise from formation of 8-OH-dG involve GC --> TA transversions. In view of these findings, OFR are considered as an important class of carcinogens. The effect of OFR is balanced by the antioxidant action of non-enzymatic antioxidants as well as antioxidant enzymes. Non-enzymatic antioxidants involve vitamin C, vitamin E, carotenoids (CAR), selenium and others. However, under certain conditions, some antioxidants can also exhibit a pro-oxidant mechanism of action. For example, beta-carotene at high concentration and with increased partial pressure of dioxygen is known to behave as a pro-oxidant. Some concerns have also been raised over the potentially deleterious transition metal ion-mediated (iron, copper) pro-oxidant effect of vitamin C. Clinical studies mapping the effect of preventive antioxidants have shown surprisingly little or no effect on cancer incidence. The epidemiological trials together with in vitro experiments suggest that the optimal approach is to reduce endogenous and exogenous sources of oxidative stress, rather than increase intake of anti-oxidants. In this review, we highlight some major achievements in the study of DNA damage caused by OFR and the role in carcinogenesis played by oxidatively damaged DNA. The protective effect of antioxidants against free radicals is also discussed.

Role of oxygen radicals in DNA damage and cancer incidence

For more than 70 years, researchers in several areas of science have been intrigued by the physical and chemical properties of the lowest excited states of molecular oxygen. With two singlet states lying close above its triplet ground state, the O2 molecule possesses a very unique configuration, which gives rise to a very rich and easily accessible chemistry, and also to a number of important photophysical interactions. In particular, photosensitized reactions of the first excited state, O2(∆g), play a key role in many natural photochemical and photobiological processes, such as photodegradation and aging processes including even photocarcinogenesis. Reactions of O2(∆g) are associated with significant applications in several fields, including organic synthesis, bleaching processes, and, most importantly, the photodynamic therapy of cancer, which has now obtained regulatory approval in most countries for the treatment of several types of tumors. The development of both applications and novel observation techniques has strongly accelerated during the past few years. Significant recent advances include, for example, the development of novel luminescent singlet oxygen probes,1-4 the time-resolved detection of O2(∆g) in a transmission microscope,5 the first time-resolved measurements of singlet oxygen luminescence in vivo,6 and the observation of oxygen quenching of triplet-excited single molecules.7 Experimental and theoretical studies on the mechanisms of photosensitized formation of excited O2 states and of their deactivation have been performed for almost 40 years. While most early liquid-phase studies were exclusively concerned with O2(∆g), recent technological advances also made possible time-resolved investigations of the second excited state, O2(Σg), which can be formed in competition with O2(∆g) in many cases. Physical mechanisms of generation and deactivation of singlet oxygen.

We introduce density functional theory and review recent progress in its application to transition metal chemistry. Topics covered include local, meta, hybrid, hybrid meta, and range-separated functionals, band theory, software, validation tests, and applications to spin states, magnetic exchange coupling, spectra, structure, reactivity, and catalysis, including molecules, clusters, nanoparticles, surfaces, and solids.

Density functional theory for transition metals and transition metal chemistry

Carotenoids are abundant in many fruits and vegetables and they play diverse roles in photobiology, photochemistry and medicine. This review concerns the reactivity of carotenoids with singlet oxygen and the interaction of carotenoids with a range of free radicals. Mechanisms associated with the anti- and pro-oxidant behaviour of carotenoids are discussed including carotenoid interactions with other anti-oxidants.

β-Carotene with vitamins E and C offers synergistic cell protection against NOx

The synthesis and photophysical study of (C/\N)Pt(II)Q complexes, where C/\N is a bidentate cyclometalating ligand and Q is 8-hydroxyquinoline or quinoline-8-thiol, are presented. The compounds were obtained as a single isomer with N atoms of the C/\N and Q ligands trans-coordinated to the Pt(II) center as shown by X-ray crystallography. These chromophores absorb intensely in the visible region and emit in the deep-red spectral region from a quinolate-centered triplet intraligand charge-transfer excited state. The emission maxima are in the range 675-740 nm, with the quantum yields and lifetimes of up to 0.82% and 5.3 mus, respectively, in deoxygenated organic solvents at room temperature. These complexes are efficient photosensitizers of singlet oxygen in air-saturated solutions, with yields up to 90%.

Pulse radiolysis studies have been used to determine the electron-transfer rate constants between various pairs of carotenoids, one of which is present as the radical cation. These dietary carotenoids include those of importance to vision, namely zeaxanthin and lutein. These results have suggested the order of relative ease of electron transfer between six carotenoids. Additional experiments, involving electron transfer between astaxanthin (ASTA), β-apo-8‘-carotenal (APO), and vitamin E (TOH), lead to the following order in terms of relative ease of electron transfer for the seven carotenoid radical cations studied:  astaxanthin > β-apo-8‘-carotenal > canthaxanthin > lutein > zeaxanthin > β-carotene > lycopene, such that lycopene is the strongest reducing agent (the most easily oxidized) and astaxanthin is the weakest, and the radical cations of the visual carotenoids, lutein (LUT) and zeaxanthin (ZEA), are reduced by lycopene (LYC) but not by β-carotene (β-CAR).
