Journal ArticleDOI
Monoclonal TCR-redirected tumor cell killing
Nathaniel Liddy,Giovanna Bossi,Katherine Adams,Anna Lissina,Tara Mahon,Namir J. Hassan,Jessie Gavarret,Frayne Bianchi,Nicholas J. Pumphrey,Kristin Ladell,Emma Gostick,Andrew K. Sewell,Nikolai Lissin,Naomi Harwood,Peter Eamon Molloy,Yi Li,Brian Cameron,Malkit Sami,Emma Baston,Penio Todorov,Samantha Paston,Rebecca Dennis,Jane Harper,Steve M. Dunn,Rebecca Ashfield,Andrew M.F. Johnson,Yvonne McGrath,Gabriela Plesa,Carl H. June,Michael Kalos,David Price,Annelise Vuidepot,Daniel Williams,Deborah H. Sutton,Bent K. Jakobsen +34 more
TL;DR: Four new reagents termed immune-mobilizing monoclonal TCRs against cancer (ImmTACs) effectively redirected T cells to kill cancer cells expressing extremely low surface epitope densities and suppressed tumor growth in vivo.
Journal ArticleDOI
An approved in vitro approach to preclinical safety and efficacy evaluation of engineered T cell receptor anti-CD3 bispecific (ImmTAC) molecules.
Jane Harper,Katherine Adams,Giovanna Bossi,Debbie E. Wright,Andrea Stacey,Nicole Bedke,Ruth Martinez-Hague,Dan Blat,Laure Humbert,Hazel Buchanan,Gabrielle S. Le Provost,Zoe Donnellan,Ricardo J. Carreira,Samantha Paston,Luise U. Weigand,Martina Canestraro,Joseph P. Sanderson,Sophie Botta Gordon-Smith,Kate L. Lowe,Karolina A. Rygiel,Alex Powlesland,Annelise Vuidepot,Namir J. Hassan,Brian Cameron,Bent K. Jakobsen,Joseph Dukes +25 more
TL;DR: This package comprises a broad range of cellular and molecular assays using human tissues and cultured cells to test efficacy, safety and specificity, and hence predict human responses in clinical trials and proposes that this entirely in vitro package offers a potential model to be applied to screening other TCR-based biologics.
Journal ArticleDOI
Examining the presentation of tumor-associated antigens on peptide-pulsed T2 cells
Giovanna Bossi,Andrew B. Gerry,Samantha Paston,Deborah H. Sutton,Namir J. Hassan,Bent K. Jakobsen +5 more
TL;DR: It is indicated that peptide concentrations in the low nanomolar range are required for T2 cells to present TAAs in extents that are comparable to those of malignant cells.
Journal ArticleDOI
Elimination of Latently HIV-infected Cells from Antiretroviral Therapy-suppressed Subjects by Engineered Immune-mobilizing T-cell Receptors.
Hongbing Yang,Sandrine Buisson,Giovanna Bossi,Zoë Wallace,Gemma Hancock,Chun So,Rebecca Ashfield,Annelise Vuidepot,Tara Mahon,Peter Eamon Molloy,Joanne Oates,Samantha Paston,Milos Aleksic,Namir J. Hassan,Bent K. Jakobsen,Lucy Dorrell +15 more
TL;DR: T-cell receptors specific for an immunodominant Gag epitope, SL9, and its escape variants were engineered to achieve supraphysiological affinity and fused to a humanised CD3-specific single chain antibody fragment, and the efficiency of infected cell elimination correlated with HIV Gag expression.
Journal ArticleDOI
TCRs with Distinct Specificity Profiles Use Different Binding Modes to Engage an Identical Peptide-HLA Complex.
C.H. Coles,R. Mulvaney,S. Malla,A. Walker,K. Smith,Angharad Lloyd,K.L. Lowe,Michelle L. McCully,R. Martinez Hague,M. Aleksic,Jane Harper,Samantha Paston,Z. Donnellan,Fiona Chester,K. Wiederhold,R.A. Robinson,Andrew Alexander Knox,A.R. Stacey,Joseph Dukes,Emma Baston,S. Griffin,Bent K. Jakobsen,Annelise Vuidepot,S. Harper +23 more
TL;DR: Data show that TCRs specific for a cognate peptide recognize discrete peptide repertoires and reconciles how an individual’s limited TCR repertoire following negative selection in the thymus is able to recognize a vastly larger antigenic pool.
