Akt-Mediated Phosphorylation of Argonaute 2 Downregulates Cleavage and Upregulates Translational Repression of MicroRNA Targets
Shane R. Horman,Maja M. Janas,Maja M. Janas,Claudia Litterst,Bingbing Wang,Bingbing Wang,Ian J. MacRae,Mary J. Sever,David V. Morrissey,Paul R. Graves,Biao Luo,Shaikamjad Umesalma,Hank H. Qi,Loren Miraglia,Carl D. Novina,Carl D. Novina,Anthony P. Orth +16 more
TL;DR: Akt3-mediated phosphorylated Argonaute 2 is a molecular switch between target mRNA cleavage and translational repression activities of Ago2, and Akt3 coimmunoprecipitates with Ago2 and phosphorylation of Ago1 facilitates its interaction with GW182 and localization to cytoplasmic processing bodies (P bodies), where miRNA-targeted mRNAs are thought to be stored and degraded.
Journal ArticleDOI
Histone demethylase PHF8 promotes epithelial to mesenchymal transition and breast tumorigenesis.
Peng Shao,Qi Liu,Peterson Kariuki Maina,Jiayue Cui,Thomas B. Bair,Tiandao Li,Shaikamjad Umesalma,Weizhou Zhang,Hank H. Qi +8 more
TL;DR: The novel functions of PHF8 in EMT and breast cancer development are reported and its candidacy as a promising therapeutic target for this disease is conferred.
Journal ArticleDOI
A porcine model of neurofibromatosis type 1 that mimics the human disease.
Katherine A. White,Vicki J. Swier,Jacob T. Cain,Jordan L. Kohlmeyer,David K. Meyerholz,Munir R. Tanas,Johanna Uthoff,Emily Hammond,Emily Hammond,Hua Li,Frank Rohret,Adam Goeken,Chun Hung Chan,Mariah R. Leidinger,Shaikamjad Umesalma,Margaret R. Wallace,Rebecca D. Dodd,Karin Panzer,Amy H. Tang,Benjamin W. Darbro,Aubin Moutal,Song Cai,Wennan Li,Shreya S. Bellampalli,Rajesh Khanna,Christopher S. Rogers,Jessica C. Sieren,Jessica C. Sieren,Dawn E. Quelle,Jill M. Weimer +29 more
TL;DR: In this paper, the authors describe a potentially novel Yucatan miniswine model bearing a heterozygotic mutation in NF1 (exon 42 deletion) orthologous to a mutation found in patients.
Journal ArticleDOI
Gene Expression Signatures Identify Novel Therapeutics for Metastatic Pancreatic Neuroendocrine Tumors
Aaron T. Scott,Michelle Weitz,Patrick Breheny,Po Hien Ear,Benjamin W. Darbro,Benjamin W. Darbro,Bartley Brown,Terry A. Braun,Guiying Li,Shaikamjad Umesalma,Courtney A. Kaemmer,Chandra K. Maharjan,Dawn E. Quelle,Dawn E. Quelle,Andrew M. Bellizzi,Andrew M. Bellizzi,Chandrikha Chandrasekharan,Chandrikha Chandrasekharan,Joseph S. Dillon,Joseph S. Dillon,Thomas M. O'Dorisio,Thomas M. O'Dorisio,James R. Howe,James R. Howe +23 more
TL;DR: The potential utility of these drugs was confirmed by in vitro cytotoxicity assays, suggesting drugs targeting MEK and TOP2A may be highly efficacious against metastatic pNETs.
Journal ArticleDOI
Epidermal Growth Factor Receptor Is Critical For Angiotensin II–Mediated Hypertrophy in Cerebral Arterioles
TL;DR: Results suggest that Ang II–induced hypertrophy in cerebral arterioles involves EGFR-dependent signaling and may include Cav-1 and nonreceptor tyrosine kinase c-Src, and is independent of blood pressure.
