The Interferon System

The family of interferon (IFN) proteins has now more than reached the potential envisioned by early discovering virologists: IFNs are not only antivirals with a spectrum of clinical effectiveness against both RNA and DNA viruses, but are also the prototypic biological response modifiers for oncology, and show effectiveness in suppressing manifestations of multiple sclerosis. Studies of IFNs have resulted in fundamental insights into cellular signalling mechanisms, gene transcription and innate and acquired immunity. Further elucidation of the multitude of IFN-induced genes, as well as drug development strategies targeting IFN production via the activation of the Toll-like receptors (TLRs), will almost certainly lead to newer and more efficacious therapeutics. Our goal is to offer a molecular and clinical perspective that will enable IFNs or their TLR agonist inducers to reach their full clinical potential.

Interferons at age 50: past, current and future impact on biomedicine.

Immobilized metal ion affinity adsorption (IMA adsorption) is a collective term that is proposed to include all kinds of adsorptions whereby metal atoms or ions immobilized on a polymer cause or dominate the interaction at the sorption sites. IMA chromatography is one of the most powerful methods available to date for protein fractionation although this is not as yet widely recognized. This study deals with the theoretical aspects of IMA adsorption and its practical applications as exemplified by the various results reported here. The synthesis of iminodiacetate-substituted agarose (IDA-agarose) and tris(carboxymethyl)ethylenediamine-agarose (TED-agarose) is described. Many types of metal ions can easily be immobilized on these gel derivatives to form IMA adsorbents. We have not observed any damage to the proteins during the adsorption-desorption process. After performance of an experiment, the gels can easily be regenerated and can be loaded with the same or a different metal ion for an ensuing experiment. Specific adsorption is demonstrated for serum proteins on immobilized Ni(II) and Fe(III). Ligand-specific desorption (affinity elution) is also demonstrated by including in the buffer system certain solutes which are similar to or identical with some particular amino acids found in proteins. High concentrations of certain salts that affect the structure of water, such as Na2SO4, promote coordinate covalent bonding of proteins by a mechanism that is apparently similar to that found in hydrophobic interactions. Neutral detergents and aquoorganic solvents may be used. This opens up the possibility for the fractionation of membrane components. The IMA-adsorption method could also be expanded to other areas besides protein fractionation.

The antigen-specific activation of T cells is dependent upon two distinct but interdependent macrophage signals. Macrophages are required to bin^, process, and present antigens in an immunogcnic form to T cells (Schwartz et ul. 1978). This function of the macrophage is genetically-restricted and is a property of an la positive subpopulation of macrophages. The second macfophage-derived T cell activating signal is a Iow molecular weight antigen-non-specific peptide termed interleukin 1 (IL I) (Aarden et al. 1979) that is apparently produced by all macrophages (Oppenheim etal. 1979). As will be discussed in this review, IL I appears to act as a maturational signal, preparing T cells to respond to antigens or secondary mediator signals. One of the major activities of interleukin 1 is to induce the synthesis and secretion of the T cell-derived mitogenic lymphokine, interleukin 2 (IL2) (Farrar et al. 1980b, Smith et al. 1980, Larsson et al. 1980). This link between IL 1 and IL 2 is an essential element in the T cell activation sequence because it involves the conversion of a primary macrophage-derived maturational signal into a secondary T cell-derived proliferative signal that results in the amplification of specific immune responses.

Interferon treatment of murine C-type oncornavirus carrier lines results in a drastic inhibition of extracellular virus formation. When such cells are subcultured in the presence of interferon, synthesis of extracellular C-type virus remains depressed until the interferon treatement is discontinued. Cell-associated virus as seen in the electron microscope or as detected by uridine incorporation is not proportionally inhibited. Several possibilities to explain this discrepancy are tested. The most likely hypothesis is that interferon treatment primarily inhibits virus release.

Purification of human fibroblast interferon by zinc chelate affinity chromatography.

Human fibroblast interferon (F-interferon) purified by adsorption on controlled-pore glass was given intramuscularly to patients at daily dosages of up to 20 × 10 6 units. Serum levels of antiviral activity were low or undetectable. In contrast, reasonably high serum titers were found in patients receiving interferon prepared from leukocytes (L-interferon). Similarly, in rabbits lower serum titers were seen with F-interferon than with L-interferon. These results are at variance with those obtained earlier (V. G. Edy, A. Billiau, and P. De Somer, J. Infect. Dis. 133: A18–A21, 1976). Possible explanations for this discrepancy are discussed. The F-interferon evoked febrile reactions, delayed skin reactivity, and transitory lymphopenia in humans. Some patients developed an allergic state of the reaginic type as evidenced by a weal and flare reaction after intradermal challenge. However, these patients did not show allergic symptoms after intramuscular injections. None of the side effects was severe enough to prohibit continuation of the treatment; most of them seemed to be due to contaminants not removed by the purification method. The possibility is considered that some of the side effects, e.g., delayed skin reactivity, are sufficiently specific to justify identification of the active principals. Images

Human Fibroblast Interferon for Clinical Trials: Pharmacokinetics and Tolerability in Experimental Animals and Humans

Summary
Human fibroblast and mouse L929 cell interferons can be purified by adsorption to and subsequent elution from Controlled Pore Glass. Purification of 40 to 90-fold to specific activities of 1 to 5 × 106 units/mg of protein can be achieved in a single step, with good recovery of activity. Human leukocyte interferon does not bind to the glass and cannot be purified in this way.
