Journal ArticleDOI

Kinetic competition during the transcription cycle results in stochastic RNA processing

TL;DR: This study uses dual-color single-molecule RNA imaging in living human cells to construct a complete kinetic profile of transcription and splicing of the β-globin gene and finds that kinetic competition results in multiple competing pathways for pre-mRNA splicing.
Journal ArticleDOI

ALS mutant FUS proteins are recruited into stress granules in induced pluripotent stem cell-derived motoneurons

TL;DR: It is shown that aberrant localization and recruitment of FUS into stress granules (SGs) is a prerogative of the FUS mutant proteins and occurs only upon induction of stress in both undifferentiated iPSCs and spinal cord neural cells.
Journal ArticleDOI

FUS Mutant Human Motoneurons Display Altered Transcriptome and microRNA Pathways with Implications for ALS Pathogenesis

TL;DR: It is reported that relevant targets of miR-375, including the neural RNA-binding protein ELAVL4 and apoptotic factors, are aberrantly increased in FUS mutant motoneurons.
Journal ArticleDOI

Background-deflection Brillouin microscopy reveals altered biomechanics of intracellular stress granules by ALS protein FUS.

TL;DR: A non-contact and label-free imaging method, named background-deflection Brillouin microscopy, is presented, enabling the first direct biomechanical analysis on intracellular stress granules containing ALS mutant FUS protein in fixed cells at a sub-micron resolution.
