Deleterious effects of climate change and human activities, as well as diverse environmental stresses, present critical challenges to food production and the maintenance of natural diversity. These challenges may be met by the development of novel crop varieties with increased biotic or abiotic resistance that enables them to thrive in marginal lands. However, considering the diverse interactions between crops and environmental factors, it is surprising that evolutionary principles have been underexploited in addressing these food and environmental challenges. Compared with domesticated cultivars, crop wild relatives (CWRs) have been challenged in natural environments for thousands of years and maintain a much higher level of genetic diversity. In this review, we highlight the significance of CWRs for crop improvement by providing examples of CWRs that have been used to increase biotic and abiotic stress resistance/tolerance and overall yield in various crop species. We also discuss the surge of advanced biotechnologies, such as next-generation sequencing technologies and omics, with particular emphasis on how they have facilitated gene discovery in CWRs. We end the review by discussing the available resources and conservation of CWRs, including the urgent need for CWR prioritization and collection to ensure continuous crop improvement for food sustainability.

Back into the Wild – Apply Untapped Genetic Diversity of Wild Relatives for Crop Improvement

Knowing pests and diseases that may cause injuries and are likely to affect plant health and quality is critical to minimizing the gap between attainable yield and actual yield. In this paper, we highlight concepts and strategies aimed at controlling major biotic constraints affecting wheat in intensive production systems and present emerging challenges, with a special attention to the developing world. Disease epidemics result from the combination of inoculum, favorable environment, and host susceptibility. Changes in cropping systems as a result of adoption of conservation agriculture may have serious implications. Necrotrophic pathogens such as those responsible for tan spot or septorias are likely to emerge, and Fusarium head blight may increase. However, resistance breeding combined with rotations, timely sowing, and irrigation or even fungicide utilization, if affordable, are part of integrated crop management practices that can minimize losses. In South Asia, the effect of spot blotch, a devastating foliar disease caused by Cochliobolus sativus, can be minimized by reducing physiological stress through timely sowing and adequate use of fertilizers, which demonstrates the complex relationships among crop physiology, disease resistance, and yield. Although some root rots that induce premature death of tillers in cooler high-yielding humid environments can be important, the dryland crown rot (Fusarium spp.), common root rot (C. sativus), and the cereal nematode (Heterodera spp. and Pratylenchus spp.) should not be ignored. These are all known to be much more damaging under suboptimal moisture (rainfed or supplementary irrigation), particularly where plant growth is stressed. Climate change is likely to modify the wheat disease spectrum in some regions, and pathogens or pests considered unimportant today may turn out to be potential new threats in future.

The challenges of maintaining wheat productivity : pests, diseases, and potential epidemics

Incorporation of genetic diversity into elite wheat (Triticum aestivum L., 2n1⁄4 6x1⁄4 42, AABBDD) cultivars has long been recognized as a means of improving wheat productivity and securing global wheat supply. Synthetic hexaploid wheat (SHW) genotypes recreated from its two progenitor species, the tetraploid, Triticum turgidum (2n1⁄4 4x1⁄4 28, AABB) and its diploid wild relative, Aegilops tauschii (2n1⁄4 2x1⁄4 14, DD) are a useful resource of new genes for hexaploidwheat improvement. These include many productivity traits such as abiotic (drought, heat, salinity/sodicity, andwaterlogging) and biotic (rusts, septoria, barley yellow dwarfvirus (BYDV), crownrot, tan spot, spot blotch, nematodes,powderymildew, and fusarium head blight) stress resistance/tolerances as well as novel grain quality traits. Numerous SHWs have been produced globally by various institutions including CIMMYT-Mexico, ICARDA-Syria, Department of Primary Industries (DPI), Victoria-Australia, IPK-Germany, Kyoto University-Japan, and USDAARS. This review examines the varied aspects in the utilization of synthetics for wheat improvement including the traits and genes identified, mapped, and transferred to common wheat. It has also been demonstrated that synthetic backcross-derived lines (SBLs, i.e., when SHW is crossed to adapted local bread varieties) show significant yield increases and thus, enhanced yield performance across a diverse range of environments, demonstrating their potential for improving wheat productivity worldwide. This is particularly evident in moisturelimited environments. The use of SBLs, advanced backcross QTL analysis, chromosome introgression lines, and whole genome association mapping is contributing to the elucidation of the genetic architecture of some of the traits. The contribution of transgressive segregation to enhanced phenotypes and the mechanisms including its genetic and physiological basis are yet to be elucidated. 36 FRANCIS C. OGBONNAYA ET AL.

Synthetic Hexaploids: Harnessing Species of the Primary Gene Pool for Wheat Improvement

Powdery mildew resistance from Thinopyrum intermedium was introgressed into common wheat (Triticum aestivum L.). Genetic analysis of the F(1), F(2), F(3) and BC(1) populations from powdery mildew resistant line CH5025 revealed that resistance was controlled by a single dominant allele. The gene responsible for powdery mildew resistance was mapped by the linkage analysis of a segregating F(2) population. The resistance gene was linked to five co-dominant genomic SSR markers (Xcfd233, Xwmc41, Xbarc11, Xgwm539 and Xwmc175) and their most likely order was Xcfd233-Xwmc41-Pm43-Xbarc11-Xgwm539-Xwmc175 at 2.6, 2.3, 4.2, 3.5 and 7.0 cM, respectively. Using the Chinese Spring nullisomic-tetrasomic and ditelosomic lines, the polymorphic markers and the resistance gene were assigned to chromosome 2DL. As no powdery mildew resistance gene was previously assigned to chromosome 2DL, this new resistance gene was designated Pm43. Pm43, together with the identified closely linked markers, could be useful in marker-assisted selection for pyramiding powdery mildew resistance genes.

Inheritance and mapping of powdery mildew resistance gene Pm43 introgressed from Thinopyrum intermedium into wheat.

Powdery mildew, caused by Blumeria graminis f. sp. tritici, is one of the most important wheat diseases worldwide in areas with cool or maritime climates. Wild emmer (Triticum turgidum var. dicoccoides) is an important potential donor of disease resistances and other traits for common wheat improvement. A powdery mildew resistance gene was transferred from wild emmer accession G-303-1M to susceptible common wheat by crossing and backcrossing, resulting in inbred line P63 (Yanda1817/G-303-1 M//3*Jing411, BC2F6). Genetic analysis of an F2 population and the F2:3 families developed from a cross of P63 and a susceptible common wheat line Xuezao showed that the powdery mildew resistance in P63 was controlled by a single recessive gene. Molecular markers and bulked segregant analysis were used to characterize and map the powdery mildew resistance gene. Nine genomic SSR markers (Xbarc7, Xbarc55, Xgwm148, Xgwm257, Xwmc35, Xwmc154, Xwmc257, Xwmc382, Xwmc477), five AFLP-derived SCAR markers (XcauG3, XcauG6, XcauG10, XcauG20, XcauG22), three EST–STS markers (BQ160080, BQ160588, BF146221) and one RFLP-derived STS marker (Xcau516) were linked to the resistance gene, designated pm42, in P63. pm42 was physically mapped on chromosome 2BS bin 0.75–0.84 using Chinese Spring nullisomic-tetrasomic, ditelosomic and deletion lines, and was estimated to be more than 30 cM proximal to Xcau516, a RFLP-derived STS marker that co-segregated with the wild emmer-derived Pm26 which should be physically located in 2BS distal bin 0.84–1.00. pm42 was highly effective against 18 of 21 differential Chinese isolates of B. graminis f. sp. tritici. The closely linked molecular markers will enable the rapid transfer of pm42 to wheat breeding populations thus adding to their genetic diversity.

Powdery mildew, caused by Blumeria graminis f sp tritici, is a widespread wheat disease in China Identification of race-specific genes and adult plant resistance (APR) is of major importance in breeding for an efficient genetic control strategy The objectives of this study were to (i) identify genes that confer seedling resistance to powdery mildew in Chinese bread wheat cultivars and introductions used by breeding programs in China and (ii) evaluate their APR in the field The results showed that (i) 98 of 192 tested wheat cultivars and lines appear to have one or more resistance genes to powdery mildew; (ii) Pm8 and Pm4b are the most common resistance genes in Chinese wheat cultivars, whereas Pm8 and Pm3d are present most frequently in wheat cultivars introduced from CIMMYT, the United States, and European countries; (iii) genotypes carrying Pm1, Pm3e, Pm5, and Pm7 were susceptible, whereas those carrying Pm12, Pm16, and Pm20 were highly resistant to almost all isolates of B graminis f sp tritici tested; and (iv) 22 genotypes expressed APR Our data showed that the area under the disease progress curve, maximum disease severity on the penultimate leaf, and the disease index are good indicators of the degree of APR in the field It may be a good choice to combine major resistance genes and APR genes in wheat breeding to obtain effective resistance to powdery mildew

Seedling and Adult Plant Resistance to Powdery Mildew in Chinese Bread Wheat Cultivars and Lines

Powdery mildew caused by Blumeria
 graminis f. sp. tritici is one of the most important wheat diseases worldwide and breeding for resistance using diversified disease resistance genes is the most promising approach to prevent outbreaks of powdery mildew. A powdery mildew resistance gene, originating from wild emmer wheat (Triticum turgidum var. dicoccoides) accessions collected from Israel, has been transferred into the hexaploid wheat line 3D232 through crossing and backcrossing. Inoculation results with 21 B.
 graminis f. sp. tritici races indicated that 3D232 is resistant to all of the powdery mildew isolates tested. Genetic analyses of 3D232 using an F2 segregating population and F3 families indicated that a single dominant gene, Ml3D232, confers resistance in the host seedling stage. By applying molecular markers and bulked segregant analysis (BSA), we have identified polymorphic simple sequence repeats (SSR), expressed sequence tags (EST) and derived sequence tagged site (STS) markers to determine that the Ml3D232 is located on chromosome 5BL bin 0.59–0.76. Comparative genetic analyses using mapped EST markers and genome sequences of rice and Brachypodium established co-linearity of the Ml3D232 genomic region with a 1.4 Mb genomic region on Brachypodium distachyon chromosome 4, and a 1.2 Mb contig located on the Oryza sativa chromosome 9. Our comparative approach enabled us to develop new EST–STS markers and to delimit the genomic region carrying Ml3D232 to a 0.8 cM segment that is collinear with a 558 kb region on B. distachyon. Eight EST markers, including an NBS-LRR analog, co-segregated with Ml3D232 to provide a target site for fine genetic mapping, chromosome landing and map-based cloning of the powdery mildew resistance gene. This newly developed common wheat germplasm provides broad-spectrum resistance to powdery mildew and a valuable resource for wheat breeding programs.

High yield potential and the wide adaptability of wheat-rye T1BL·1RS translocation lines are attractive to breeders. The wheat-rye lines Lankao 1, 3, 4, and 5 were resistant to a wide spectrum of wheat powdery mildew (Blumeria graminis f. sp. tritici) isolates from both China and Canada. They also were resistant to a mixture of wheat stem rust (Puccinia graminis f. sp. tritici) pathotypes (98WSR) and wheat stripe rust (P. striiformis f. sp. tritici) races from western Canada and China. Colonization of wheat curl mite (WCM) (Aceria tosichella) resulted in slower development of rolling and trapping leaves in the Lankao lines than in the WCM-susceptible check cultivars. The delayed development of Wheat streak mosaic (WSM) symptoms on Lankao lines was observed when transmitted by viruliferous WCM, even though they were susceptible to Wheat streak mosaic virus (WSMV). Characterization of Wheat-Triticale Lines Resistant to Powdery Mildew, Stem Rust, Stripe Rust, Wheat Curl Mite, and Limitation on Spread of WSMV.

Field and controlled environmental tests indicated that the 49 accessions of closely related species and 12 landraces of wheat (Triticum aestivum L. em. Thell.) from the National Gene Bank of China showed different reactions to powdery mildew (Blumeria graminis (DC.) E. O. Speer. f. sp. tritici) and stripe rust (Puccinia striiformis Westend f. sp. tritici) at adult and seedling stages. Unknown Pm genes or alleles were postulated with Triticum baeoticum Boiss. accessions BO 3 and Triticum monococcum L. MO 4 and MO 5 when inoculated with 21 powdery mildew isolates at seedling stage. Fourteen accessions of T. baeoticum, T. monococcum,Triticum durum, and wheat landraces were inoculated with 30 stripe rust isolates at seedling stage. Unknown Yr genes or alleles were postulated with T. baeoticum Boiss. accession BO 5, as well as wheat landraces Xiaobaimai, Laomangmai, and Shaanxibai. Heterogeniety in reaction to powdery mildew isolates and stripe rust races were observed in related species and landraces of wheat.
