■ Abstract Apoptosis is a genetically programmed, morphologically distinct form of cell death that can be triggered by a variety of physiological and pathological stimuli. Studies performed over the past 10 years have demonstrated that proteases play critical roles in initiation and execution of this process. The caspases, a family of cysteine-dependent aspartate-directed proteases, are prominent among the death proteases. Caspases are synthesized as relatively inactive zymogens that become activated by scaffold-mediated transactivation or by cleavage via upstream proteases in an intracellular cascade. Regulation of caspase activation and activity occurs at several different levels: ( a) Zymogen gene transcription is regulated; ( b) antiapoptotic members of the Bcl-2 family and other cellular polypeptides block proximity-induced activation of certain procaspases; and ( c) certain cellular inhibitor of apoptosis proteins (cIAPs) can bind to and inhibit active caspases. Once activated, caspases cleave a variety of intracellular polypeptides, including major structural elements of the cytoplasm and nucleus, components of the DNA repair machinery, and a number of protein kinases. Collectively, these scissions disrupt survival pathways and disassemble important architectural components of the cell, contributing to the stereotypic morphological and biochemical changes that characterize apoptotic cell death.

Current approaches and challenges for the metabolite profiling of complex natural extracts

Incarvillea compacta Maxim. has been used to treat stomach disease in Tibet for many years. The objectives of this study were to explore the anti-cancer ability of trichloromethane fraction of I. compacta Maxim. roots (IC-TCL, R2) in EBV positive AGS cancer cells and its effects on cell cycle arrest, apoptosis and lytic induction. MTT and trypan blue assays were to detect the inhibitory effects of different fraction in different cell lines. Hoechst 33342 staining, Annexin V-PE/7-AAD staining and DIOC6 staining were used to detect the apoptosis induction effects of R2. Western blot experiments were used to detect the expression of apoptosis related proteins BAX and Bcl-2, EBV lytic related proteins BZLF1 and BMRF1, cell cycle regulation related proteins Cyclin D1 and RB after R2 treatment. Cell cycle arrest was analyzed by flow cytometry. MTT and trypan blue assays revealed that R2 could significantly reduce cell viability in a dose-dependent manner in EBV positive AGS cells compared with non-EBV infected AGS and other cancer cell lines, whereas n-BuOH and H2O fractions showed non-inhibitory effects in tested cancer cells. R2 could decrease mitochondrial membrane potential and the expression of Bcl-2, while increase the expression of BAX. R2 could also induce EBV lytic replication by activating mRNA levels of BZLF1, BRLF1 and BMRF1. Protein expressions of BZLF1 and BMRF1 were also increased after R2 treatment. Cell cycle analysis showed that R2 treatment could induce G0/G1 phase arrest. The expression of Cyclin D1 decreased, while Rb increased. These results demonstrated that R2 could inhibit the proliferation of AGS-EBV cancer cells by inducing EBV lytic replication, apoptosis and G0/G1 arrest, through the regulation of related proteins. Therefore, R2 could be used as a potential treatment in AGS-EBV cells.

Trichloromethane fraction of Incarvillea compacta induces lytic cytotoxicity and apoptosis in Epstein-Barr virus-positive gastric cancer AGS cells

A series of novel oleanolic acid 3-O-β-d-glucuronopyranoside derivatives have been designed and synthesized. Biological evaluation has indicated that some of the synthesized compounds exhibit moderate to good activity against H₂O₂-induced injury in rat myocardial cells (H9c2). Particularly, derivative 28-N-isobutyl ursolic amide 3-O-β-d-galactopyranoside (8a) exhibited a greater protective effect than the positive control oleanolic acid 3-O-β-d-glucuronopyranoside, indicating that it possesses a great potential for further development as a cardiovascular disease modulator by structural modification.

Soulieoside R : A New Cycloartane Triterpenoid Glycoside from Souliea vaginata

The genus Maytenus is a member of the Celastraceae family, of which several species have long been used in traditional medicine. Between 1976 and 2021, nearly 270 new compounds have been isolated and elucidated from the genus Maytenus. Among these, maytansine and its homologues are extremely rare in nature. Owing to its unique skeleton and remarkable bioactivities, maytansine has attracted many synthetic endeavors in order to construct its core structure. In this paper, the current status of the past 45 years of research on Maytenus, with respect to its chemical and biological activities are discussed. The chemical research includes its structural classification into triterpenoids, sesquiterpenes and alkaloids, along with several chemical synthesis methods of maytansine or maytansine fragments. The biological activity research includes activities, such as anti-tumor, anti-bacterial and anti-inflammatory activities, as well as HIV inhibition, which can provide a theoretical basis for the better development and utilization of the Maytenus.
