Journal ArticleDOI

Hollow Cone Electron Imaging for Single Particle 3D Reconstruction of Proteins

TL;DR: Hollow cone dark field imaging using thermal diffuse scattered electrons gives about a 4 times contrast increase as compared to bright field imaging, and demonstrates the 3D reconstruction of a stained GroEL particle can yield about 13.5 Å resolution but using a strongly reduced number of images.
Journal ArticleDOI

Functional Studies of ssDNA Binding Ability of MarR Family Protein TcaR from Staphylococcus epidermidis.

TL;DR: It is shown that TcaR can interact strongly with single-stranded DNA (ssDNA) and preferentially binds 33-mer ssDNA over double-strand DNA and inhibits viral ssDNA replication, suggesting a novel role in regulation of DNA replication.
Journal ArticleDOI

The Arginine Pairs and C-Termini of the Sso7c4 from Sulfolobus solfataricus Participate in Binding and Bending DNA.

TL;DR: A model of the Sso7c4-DNA complex using a curved DNA molecule in the catabolite activator protein- DNA complex is proposed and a functional role for Sso 7c4 in the organization of chromosomal DNA and/or the regulation of gene expression through bridging and bending interactions is suggested.
ComponentDOI

Use of Cryo-EM To Uncover Structural Bases of pH Effect and Cofactor Bispecificity of Ketol-Acid Reductoisomerase.

TL;DR: The ketol-acid reductoisomerase (KARI) catalyzes conversion of (2S)-acetolactate or ( 2S)- acetolactates to keto-EM, where the latter is converted to polymethine by the KARI machinery.
