Journal ArticleDOI
Structural and mechanistic studies on 2-oxoglutarate-dependent oxygenases and related enzymes.
TL;DR: Crystallographic and mechanistic studies have helped to define the overall fold of the 2-oxoglutarate-dependent enzymes and have led to the identification of coordination chemistry closely related to that of other nonheme-Fe(II)-dependent oxygenases, suggesting related mechanisms for dioxygen activation that involve iron-mediated electron transfer.
Journal ArticleDOI
Structural origins of the selectivity of the trifunctional oxygenase clavaminic acid synthase.
Zhihong Zhang,Jingshan Ren,David K. Stammers,Jack E. Baldwin,Karl Harlos,Christopher J. Schofield +5 more
TL;DR: In this paper, the crystal structures of Clavaminate synthase (CAS) are described in complex with Fe(II), 2-oxoglutarate (2OG) and substrates (N-alpha-acetyl-Larginine and proclavaminic acid).
Journal ArticleDOI
Crystal structure of a clavaminate synthase–Fe(II)–2-oxoglutarate–substrate–NO complex: evidence for metal centred rearrangements
Zhihong Zhang,Jingshan Ren,Karl Harlos,Colin H McKinnon,Ian J. Clifton,Christopher J. Schofield +5 more
TL;DR: Crystals of CAS complexed with Fe(II), 2OG and deoxyguanidinoproclavaminate were exposed to nitric oxide (NO) acting as a dioxygen analogue to catalyses three steps in the biosynthesis of clavulanic acid.
Journal ArticleDOI
A procedure for setting up high-throughput nanolitre crystallization experiments. II. Crystallization results

Sleeman,Guillaume Stewart-Jones,M. Vuong,J. Welburn,Zhihong Zhang,David K. Stammers,Raymond J. Owens,E Y Jones,Karl Harlos,David I. Stuart +33 more
TL;DR: An initial tranche of results from day-to-day use of a robotic system for setting up 100 nl-scale vapour-diffusion sitting-drop protein crystallizations has been surveyed in this paper.
Journal ArticleDOI
Oxygenase-catalyzed ribosome hydroxylation occurs in prokaryotes and humans.

Coleman,Christopher J. Schofield +31 more
TL;DR: This work reveals new therapeutic possibilities via oxygenase inhibition and by targeting modified over unmodified ribosomes through inhibition of oxygenase catalyzing arginyl hydroxylation of the ribosomal protein Rpl16.
