Abstract: Histological and histochemical alterations on gill primary lamellae of the teleost, young yellowtail (Seriola quinqueradiata), affected by the sea bloom (Chattonella antiqua) were studied and causative factors responsible for fish death were considered in the present work. A significant loss of mucous goblet cells on the afferent ridges occurred within about one hr. Among the remainder, mucous cells located on the afferent ridge of the basal part of the gill primary lamellae were markedly impaired (64% decrease in number). For TEM preparation, hyaline degenerations of the mucous cell membrane were observed. Histochemically, these mucous goblet cells contained neutral and acid glycoproteins. The cell layer on both ridges exposed to sea bloom appeared to be thinner than that of the control. The cell body of the internal multilayered mass was shrunk and intercellular spaces were markedly expanded. These edematous gill lamellae might be caused by the disappearance of the mucous coat, leading to locally impaired osmoregulations. As a result, gas exchange on the gill lamellae might be disturbed.

Abstract: Resolution of human alpha-fetoprotein (AFP) into four distinct molecular species was demonstrated by a combination of two affinity chromatographies with crossed-immuno-affino-electrophoresis (CIAE) using concanavalin A (Con A) and Lens culinaris hemagglutinin (LcH)-A and LcH-B as affinity media. Of the four AFPs, AFP1 had no affinity for Con A, LcH-A, or LcH-B; AFP2 showed a high affinity for Con A, a low affinity for LcH-A, and an intermediate affinity for LcH-B (or a low affinity, depending on the lot of LcH-B preparations used); AFP3 revealed strong affinities for all of the three lectins; and AFP4, a trace component of hepatoma AFP in the present study, showed no interaction with Con A, but a definite interaction with LcH-A or LcH-B. These results were based on the determination of dissociation constants (Kd) of AFP-lectin complex by CIAE on isolated preparations of the three major hepatoma AFPs. These AFPs had identical electrophoretic mobilities of 0.86-0.87 (relative to human albumin) in the absence of lectins. The calculated mobilities of AFP2 and AFP3 were both reduced to 0.50-0.58 by saturation with lectins, but these two AFPs were clearly separated by 1 mg/ml LcH-A or LcH-B because of their large differences in Kd.

Abstract: Pollen grains of five species and two interspecific hybrids ofTrillium from eastern Asia have been examined with scanning electron and transmission electron microscopy. All species from eastern Asia, excludingT. govanianum, are characterized by inaperturate pollen and an unstratified and granulated exine structure. Pollen of the Himalayan species,T. govanianum, is, however, monocolpate and intectate. The palynological evidence suggests that the Asiatic species, excludingT. govanianum, are closely related to each other and toT. erectum in North America, andT. govanianum is independent of the other species in the genus.

Abstract: An endo-l,5-α-L-arabinase [EC 3.2.1.99] has been highly purified from the culture fluid of Bacillus subtilis IFO 3022 grown on a medium containing beet pulp extract. The molecular weight of the enzyme was 33,000 by SDS polyacrylamide gel electrophoresis. The purified enzyme was active on 1,5-arabinan, and inactive on p-nitrophenyl α-L-arabinofuranoside. The optimum pH was 6.0. The enzyme was inhibited by Hg2 + , but not by l-arabonic-γ-lactone. The purified enzyme was able to loosen potato tissue.

Abstract: The cause of red tides appearing in neritic waters in Japan have been discussed by several workers in relation to the development of eutrophication. The author has been engaged in marine environmental studies on outbreaks of red tides, mainly, in the Seto Inland Sea since 1964 using a combination of the following approaches: The red tide organisms surveyed wereEutreptiella sp. (Euglenophyceae),Dictyocha fibura (Chrysophyceae),Chattonella marina, C. antiqua, Fibrocapsa japonica, Heterosigma akashiwo (Chloromonadophyceae),Prorocentrum minimum, P. triestinum, Gymnodinium mikimotoi, G. nelsoni, Gymnodinium type '65,Noctiluca miliaris, Protogonyaulax catenella, Gonyaulax polygramma, Peridinium quinquecorne (Dinophyceae) andMesodinium rubrum (Ciliata). Most of these species were isolated and cultured for physiological studies. Algal growth potential in sea water was assayed in Hiuchi Nada and Beppu Bay. Growth promoting activity was found in sea water about 2km off shore from the industrial zone which locates along the coast of Beppu Bay. The effects of industrial wastes were examined forEutreptiella sp.,Heterosigma akashiwo andProrocentrum triestinum. Sulfite pulp waste caused marked promotion of growth in these organisms. The growth promoting activity of the waste was elucidated by chelation of the waste with iron. After the oil spill which occurred at Mizushima Bay in 1974, a heavy red tide ofNoctiluca miliaris appeared, followed byProrocentrum minimum. Polycyclic and polar fractions in heavy oil separated by chromatography were suspected to be the growth promoting substances. Natural growth promoting chemicals, uracil and thymine, were found in interstitial water of bottom mud of Beppu Bay in concentrations of 58–120, and 28–83ώg per 100 g dry mud, respectively. The mechanism of fish kills caused by red tides was another subject of study.

Abstract: The massculture of marine phytoflagellates such as Protogonyaulax catenella, Prorocentrum minimum and Chattonella antiqua in 10 l or 100 l glass tanks have been examined. Filtered sea water or commercial sea salt solution, fortified with nutrients, vitamins and EDTA-Fe and -Mn was used as the culture medium after filter sterilization. Culture temperatures are 20–23°C and light intensity supplied by white fluorescent lamps was about 5000 lux under conditions of 14 hrs. light and 10 hrs. dark. The 100 l tank was equipped with a pH controller supplying CO 2 gas to keep the pH of the medium from 8.2 to 8.7. In the latter period of the exponential phase of the cell growth, half or three quarters of the culture was harvested and fresh medium was supplied aseptically. The cell yield of P.catenella was 33.2 g(dry basis) in 39 days with 400 l . The total PSP toxicities were 329,000 MU; of P.minimum , 32 g in 33 days with 410 l media; of C.antiqua , 1.2×10 10 cells corresponding to 194 g on wet basis in 42 days with 440 l . These materials are now being used in chemical analyses.

Abstract: 3種のリン脂質のエマルションの乳化安定性に対する効果を詳しく調べ,また各分離相へのリン脂質の移行について検討し,次の結果が得られた。(1) サラダ油またはケロシンー水系エマルションの安定性は,リン脂質含量及びリン脂質組成により異なった。(2) 3種のリン脂質 (PC, PE及びPI)の乳化安定性はPI>PE>PCの順に高かった。(3) PC-PE 及びPC-PI系では, PE及びPI含量が増加するほど安定性は高く,特に, PC-PI系ではPIが約半量含まれることにより安定性はPI単純系の場合とほぼ等しかった。PE-PI系の安定性は,組成により変わらなかった。(4) 実験を行なった3成分系の中では, PC-PE-PI=1:1:2が最も安定性が高かった。(5) 界面張力が低いリン脂質によるエマルションほどクリーミング安定性は高く,また, PCの界面張力の値の経時的な低下はなだらかで,界面への吸着がかなり遅いことが推察された。(6) エマルションのクリーミングが進むと少量のリン脂質が水相へ移行し,さらに,エマルションの破壊が進み油相が分離してくると大部分が油相に移行した。

Abstract: Electroimmunoprecipitates in agarose gels were amplified with protein A-horseradish peroxidase (HRP) conjugate. The method is simple and detected quantitatively as low as 0.5 ng of human α-fetoprotein (AFP) in electroimmunodiffusion. The alternative methods employing the second antibody, labeled initially or afterwards with HRP, failed to stain AFP immunoprecipitates. Direct application of anti-AFP immunoglobulins-HRP in electroimmunodiffusion produced less contrasted colored precipitates. Immunoprecipitates of other proteins were similarly visualized by the protein A-HRP method, avoiding the use of specifically labeled antibody.

Abstract: The nutritive value of leaf protein concentrates (LPCs) made from different crops was examined in chicks in connection with methionine supplementation. Two feeding experi-ments were made in which LPCs were used as a sole source of dietary protein to provide 18% crude protein. In the first experiment, the supplementation of 0.2% L-methionine improved the nutritive value of both LPCs made from broad bean and ladino clover though there was no difference in the nutritive value between both the LPCs. In the second ex-periment, the nutritive value of five LPCs was compared with that of soybean meal or isolated soybean protein. Without the supplementary methionine, the nutritive value of LPCs made from Italian ryegrass and oats-annual blue grass mixture (3:7 in a dry matter basis) was higher than that of the leguminous LPCs made from broad bean, ladino clover and red clover. This difference was related to the sulfur-containing amino acids content in the LPC. Thus, the supplementation of 0.2% L-methionine was more effective in improving the nutritive value of the leguminous LPCs low in the sulfur-containing amino acids and consequently diminished the nutritive difference among LPCs except the broad bean LPC. However, the nutritive value of LPCs with supplemental methionine was still inferior to that of soybean meal or isolated soybean protein. The nutritive value of the alfalfa LPC was examined in the second experiment, but feeding the alfalfa LPC diet caused the death of all the chicks, regardless of the methionine supplementation.

Abstract: Pressure shifts of the hyperfine splitting in Na 32S1/2 that are due to buffer gases have been measured for the first time to our knowledge by all-optical means using the method of synchronized-quantum-beat spectroscopy reported previously [Y. Fukuda et al., Opt. Commun. 38, 357 (1981)]. The substate coherence is excited and detected with amplitude-modulated mode-locked laser pulses tuned to the D1 line (pulse-repetition frequency, fp; modulation frequency, fm), and the resonances observed at f0 = nfp + n′fm (n and n′ are integers) are used for the measurement. The pressure-shift coefficients obtained at 210°C are 82, 61, −1, and −47 Hz/Torr for He, N2, Ar, and Kr, respectively, within an accuracy of ±3 Hz/Torr.

Abstract: A plasmid, pNaDHI, carrying the nad gene for the de novo synthesis of NAD was constructed with pBR 322 and chromosomal DNA of Escherichia coli. Cleavage by restriction endonucleases showed that the plasmid contained a DNA insert of 8.9 K bp at the Hind III site of pBR322. The cell-free extract of a transformant of E. coli C600 which contained the plasmid formed 5 times more quinolinic acid from aspartic acid and dihydroxyacetone phosphate than that of C600 did. Enzyme synthesis of quinolinic acid from aspartic acid using partially of Protein B of quinolinate synthetase increased in the transformant, suggesting that the nad B gene was cloned in the transformant.

Abstract: The method of Synchronized Quantum Beat Spectroscopy has been improved by introducing the amplitude modulation on the excitation pulse train. By using an amplitude-modulated train of picosecond pulses from a cw mode-locked dye laser, we obtain resonance enhancement of the substate coherence (quantum beat) at f 0 = n f p + n ' f m ( n , n ' : integer, f 0 : sublevel splitting, f p : pulse repetition frequency, f m : modulation frequency), which can be detected by monitoring the optical anisotropy due to the substate coherence. Resonances can be obtained in the frequency domain by sweeping f m . Pressure shift coefficients of the hyperfine splitting in Na 3 2 S 1/2 are measured to be 82, 61, -1 and -47 Hz/Torr for He, N 2 , Ar and Kr, respectively, within an accuracy of ±3 Hz/Torr.

Abstract: Intense stimulated emission from sodium vapor is observed at around 800 nm when it is pumped by a pulsed dye laser around 600 nm. The spectral lines are localized around 800 nm. To investigate the origin of this emission, we try to assign the stimulated lines excited by a dye laser at 632.8 nm (the wavelength of the He-Ne laser) and by a ruby laser at 694.3 nm. Moreover, the A Σ -X Σ transitions at 595.728 and 594.664 nm of a Na 2 which are newly assigned by using the polarization labelling spectroscopy are excited by the output from a cw-dye-laser amplified with N 2 laser-pumped dye amplifier, and some of the stimulated lines are assigned. It is then concluded that the origin of these stimulated emission lines is the A Σ -X Σ transition. Also it can be explained that these stimulated lines come from around the band edge of the A Σ -X Σ transition.
