Abstract: The turnover of nicotinic acetylcholine receptors (AChR) is a critical factor that determines function and safety of neuromuscular transmission at the nerve-muscle synapses, ie neuromuscular junctions (NMJs) Previously, three different populations of AChRs exhibiting distinct stereotypic and activity-dependent half-life values were observed in mouse muscles To address AChR turnover in more detail, we here employed a recently developed longitudinal radioiodine assay that is based on repetitive measurements of radio emission from the same animals over long periods of time in combination with systematic variation of the time elapsed between AChR pulse-labeling and muscle denervation Modeling of the data revealed profiles of AChR de novo synthesis and receptor incorporation into the postsynaptic membrane Furthermore, decay of pre-existing AChRs upon denervation showed a peculiar pattern corroborating earlier findings of a two-step stabilization of AChRs

Abstract: A mathematical model of leukaemia therapy based on the Gompertzian law of cell growth is investigated. The effect of the medicine on the leukaemia and normal cells is described in terms of therapy functions. A feedback control problem with the purpose of minimizing the number of the leukaemia cells while retaining as much as possible the number of normal cells is considered. This problem is reduced to solving the nonlinear Hamilton–Jacobi–Bellman partial differential equation. The feedback control synthesis is obtained by constructing an exact analytical solution to the corresponding Hamilton–Jacobi–Bellman equation.

Abstract: Breast cancer is a heterogeneous genetic disease driven by the accumulation of individual mutations per tumor. Whole-genome sequencing approaches have identified numerous genes with recurrent mutations in primary tumors. Although mutations in well characterized tumor suppressors and oncogenes are overrepresented in these sets, the majority of the genetically altered genes have so far unknown roles in breast cancer progression. To improve the basic understanding of the complex disease breast cancer and to potentially identify novel drug targets or regulators of known cancer-driving pathways, we analyzed 86 wild-type genes and 94 mutated variants for their effect on cell growth using a serially constructed panel of MCF7 cell lines. We demonstrate in subsequent experiments that the metal cation transporter CNNM4 regulates growth by induction of apoptosis and identified a tumor suppressive role of complement factor properdin (CFP) in vitro and in vivo. CFP appears to induce the intracellular upregulation of the pro-apoptotic transcription factor DDIT3 which is associated with endoplasmic reticulum-stress response.

Abstract: Even more than 50 years after the ban of DDT in Germany, farmers are still affected by its persistence in contaminated soils. Depending on the crop cultivated on such soils, this often leads to low-level residues of DDT and its metabolites DDE and DDD (“DDX”), which are perceived as a risk by the food value chain. Pesticide formulations used in modern agriculture commonly contain high levels of surfactants, but so far no open-field studies have evaluated the effects of these treatments on the mobility of lipophilic contaminants, such as DDX. In this field trial, a 1.03 ha section was cultivated with Cucurbita maxima under realistic conditions to monitor the mobility of DDX in low-level contaminated agricultural soils in dependence of common pesticide applications.
