Abstract: Due to reduced thermal conductivity, vacuum insulation panels (VIPs) provide significant thermal insulation performance. Our novel vacuum panels operate at reduced pressure and are filled with a powder of precipitated silicic acid to further hinder convection and provide static stability against atmospheric pressure. To obtain an in depth understanding of heat transfer mechanisms, their interactions and their dependencies inside VIPs, detailed microscale simulations are conducted. Particle characteristics for silica are used with a discrete element method (DEM) simulation, using open source software Yade-DEM, to generate a periodic compressed packing of precipitated silicic acid particles. This aggregate packing is then imported into OpenLB (openlb.net) as a fully resolved geometry, and used to study the effects on heat transfer at the microscale. A three dimensional Lattice Boltzmann method (LBM) for conjugated heat transfer is implemented with open source software OpenLB, which is extended to include radiative heat transport. The infrared intensity distribution is solved and coupled with the temperature through the emissivity, absorption and scattering of the studied media using the radiative transfer equation by means of LBM. This new holistic approach provides a distinct advantage over similar porous media approaches by providing direct control and tuning of particle packing characteristics such as aggregate size, shape and pore size distributions and studying their influence directly on conduction and radiation independently. Our aim is to generate one holistic tool which can be used to generate silica geometry and then simulate automatically the thermal conductivity through the generated geometry.

Abstract: By mediating voluntary muscle movement, vertebrate neuromuscular junctions (NMJ) play an extraordinarily important role in physiology. While the significance of the nerve-muscle connectivity was already conceived almost 2000 years back, the precise cell and molecular biology of the NMJ have been revealed in a series of fascinating research activities that started around 180 years ago and that continues. In all this time, NMJ research has led to fundamentally new concepts of cell biology, and has triggered groundbreaking advancements in technologies. This review tries to sketch major lines of thought and concepts on NMJ in their historical perspective, in particular with respect to anatomy, function, and molecular components. Furthermore, along these lines, it emphasizes the mutual benefit between science and technology, where one drives the other. Finally, we speculate on potential major future directions for studies on NMJ in these fields.

Abstract: In response to gonadectomy certain inbred mouse strains develop sex steroidogenic adrenocortical neoplasms. One of the hallmarks of neoplastic transformation is expression of GATA4, a transcription factor normally present in gonadal but not adrenal steroidogenic cells of the adult mouse. To show that GATA4 directly modulates adrenocortical tumorigenesis and is not merely a marker of gonadal-like differentiation in the neoplasms, we studied mice with germline or conditional loss-of-function mutations in the Gata4 gene. Germline Gata4 haploinsufficiency was associated with attenuated tumor growth and reduced expression of sex steroidogenic genes in the adrenal glands of ovariectomized B6D2F1 and B6AF1 mice. At 12 months after ovariectomy, wild-type B6D2F1 mice had biochemical and histological evidence of adrenocortical estrogen production, whereas Gata4+/− B6D2F1 mice did not. Germline Gata4 haploinsufficiency exacerbated the secondary phenotype of postovariectomy obesity in B6D2F1 mice, presumably by limiting ectopic estrogen production in the adrenal glands. Amhr2-cre-mediated deletion of floxed Gata4 (Gata4F) in nascent adrenocortical neoplasms of ovariectomized B6.129 mice reduced tumor growth and the expression of gonadal-like markers in a Gata4F dose-dependent manner. We conclude that GATA4 is a key modifier of gonadectomy-induced adrenocortical neoplasia, postovariectomy obesity, and sex steroidogenic cell differentiation.

Abstract: Summary Ichthyoses are a group of rare genetic skin disorders that pose numerous clinical challenges, in particular with respect to the correct diagnosis and appropriate management. The present update of the German ichthyosis guidelines addresses recent diagnostic advances that have resulted in the Soreze consensus classification. In this context, we provide an updated diagnostic algorithm, taking into account clinical features as well as the molecular genetic basis of these disorders. Moreover, we highlight current therapeutic approaches such as psychosocial support, balneotherapy, mechanical scale removal, topical therapy, and systemic retinoid therapy. General aspects such as the indication for physical therapy, ergotherapy, or genetic counseling are also discussed. The present update was consented by an interdisciplinary consensus conference that included dermatologists, pediatricians, human geneticists, and natural scientists as well as representatives of the German patient support organization Selbsthilfe Ichthyose e. V.
