Detection of potato leafroll virus in single aphids by the reverse transcription polymerase chain reaction and its potential epidemiological application.

TLDR
A reverse transcription and polymerase chain reaction (RT-PCR) system was developed using two 20-mer primers located in the potato leafroll virus (PLRV) capsid gene and a 336-bp PCR product was detected from aphids which had been fed on PLRV-infected plants.

Aphid-transmitted potato viruses: The importance of understanding vector biology

TL;DR: This review addresses some of the biological complexities presented by aphid-transmitted viruses of potato, Solanum tuberosum L., and their vectors and discusses the application of this knowledge to the management of potato viruses with particular emphasis on seed potato production.

A novel usage of random primers for multiplex RT-PCR detection of virus and viroid in aphids, leaves, and tubers.

TL;DR: Comparison of RP primed cDNAs from dormant or sprouted tubers and leaves showed that for some cultivars, such as 'Shepody', leaves were more reliable for PVY and PLRV detection than the tubers, in both the d- and m-RT-PCR.

Simultaneous detection and typing of plum pox potyvirus (PPV) isolates by heminested-PCR and PCR-ELISA

TL;DR: Testing of 228 PPV-infected fruit tree samples indicated that so far only PPV type D appears to be present in Spain and in Chile, and increased sensitivity provided by heminested-PCR allowed the detection of PPV targets of D and M types, in wingless individuals of the aphid vector Aphis gossypii.

Epidemiology and Field Control of PVY and PLRV

TL;DR: Hane and Hamm as discussed by the authors showed that primary infection with potato leafroll virus (PLRV) can affect potato quality by causing phloem net necrosis, a darkening of the vascular bundle that becomes more pronounced during storage.
Journal ArticleDOI

Detection of stylet-borne and circulative potato viruses in aphids by duplex reverse transcription polymerase chain reaction.

TL;DR: Significant difference in virus detection was observed whether the aphids were tested immediately after exposure to virus sources or stored for up to 45 days in ethanol at room temperature.
