The requirement for maintenance of a background level of estrogen suggests that the inhibitory action of progesterone is mediated through progesterone receptor interactions.
In others, on the contrary, progesterone reinforces the action of estrogen; this synergistic action is found in the guinea pig, in the rat, in the mouse, and in the golden hamster.
The results indicate a critical temporal reliance of the inhibitory effects of progesterone on estrogen receptor activity and estrogen function in a well defined animal model.
The results demonstrate a tissue-specific inhibitory action of progesterone on estrogen-induced enhancement of nuclear estrogen receptor binding.
This “facilitating” effect of progesterone is thought to act synergistically with the initial “priming” effect of estrogen.
Our results (1) show that in our culture system estradiol induced up regulation of estrogen receptor and progesterone receptor A and B and suggest that the estrogen induced up regulation is prevented by progesterone (2) a clear cut antigestagenic effect of RU 486 and (3) suggest that both progesterone isoforms are analogously regulated in our culture model.
These results suggest a mechanism for antagonistic effects of estrogen and progesterone.
